中文摘要: 为探究新食品原料蓝莓花色苷的化学成分组成和体内抗氧化能力,该研究利用高效液相色谱-紫外检测器-电雾式检测器和超高液相色谱-四极杆-飞行时间质谱法对蓝莓花色苷样品进行全面表征和结构鉴定,并利用D-半乳糖致衰老模型评价其体内抗氧化活性。研究结果表明:不同批次的蓝莓花色苷化学成分一致性较好;结合保留时间、精确分子量和质谱裂解规律,从蓝莓花色苷中共鉴定出26种化合物,包括15种花色苷、9种黄酮和2种酚酸;除高含量的花色苷成分外,还存在部分含量较高的黄酮类、酚酸类和多糖类成分。灌胃蓝莓花色苷可有效提高衰老小鼠的抗氧化能力,与D-半乳糖模型组相比,蓝莓花色苷在90 mg/(kg·d)剂量下可提高血清中的总抗氧化能力、丙二醛含量、超氧化物歧化酶及谷胱甘肽过氧化物酶活力。
Abstract:To investigate the chemical composition and in vivo antioxidant capacity of blueberry anthocyanins(a novel food raw material),high performance liquid chromatography-ultraviolet detector-charged aerosol detector and ultra-high performance liquid chromatography-quadrupole-time of flight-mass spectrometry were used to comprehensively characterize and identify the structure of blueberry anthocyanin samples. Additionally,the in vivo antioxidant activity was evaluated using a D-galactose-induced aging model. The results showed that the chemical composition consistency of blueberry anthocyanins from different batches was good.A total of 26 compounds,including 15 anthocyanins,9 flavonoids,and 2 phenolic acids,were identified based on retention time,accurate molecular weight,and mass spectrometric fragmentation patterns. In addition to high-content anthocyanins,flavonoids,phenolic acids,and polysaccharides with relatively high contents were also present. Intragastric administration of blueberry anthocyanins effectively improved the antioxidant capacity of aging mice. Compared with the D-galactose model group,blueberry anthocyanins at a dose of 90 mg/(kg·d) increased the total antioxidant capacity,malondialdehyde content,superoxide dismutase activity,and glutathione peroxidase activity in serum.
