中文摘要: 为研究显齿蛇葡萄叶发酵过程中微生物群落分布和化学成分的变化,以发酵的显齿蛇葡萄叶为样品,进行微生物DNA抽提、聚合酶链式反应(polymerase chain reaction,PCR)扩增、测序文库的构建,高通量测序数据分析,鉴定微生物群落分布;在黄酮、多酚、蛋白质、氨基酸、可溶性糖和水浸出物含量测定的基础上,采用超高效液相色谱-串联质谱(ultra performance liquid chromatography-mass spectrometry,UPLC-MS/MS)法测定发酵过程中黄酮和多酚化合物的变化;采用全二维气相色谱-飞行时间质谱联用分析发酵前后挥发成分的变化。结果表明,显齿蛇葡萄叶发酵过程中,在属水平上,微生物优势群落为乳杆菌属、片球菌属和双歧杆菌属;发酵过程中,黄酮、多酚、可溶性糖和水浸出物含量在60h达到最大,二氢杨梅素、杨梅苷、花旗松素、杨梅素、槲皮苷5种黄酮化合物含量变化较小,稳定性较强,表没食子儿茶素没食子酸酯、没食子儿茶素、表没食子儿茶素等8种多酚化合物中,表没食子儿茶素没食子酸酯含量降低,表没食子儿茶素含量升高,其他多酚化合物含量基本不变,蛋白质和氨基酸含量在整个发酵期间逐渐降低;发酵过程产生的主要挥发成分有萜烯类、醇类、醛类、酯类和酮类化合物,特征香气为木香、花香、薄荷香,重要的香气成分有水杨酸甲酯、石竹烯。
Abstract:To investigate the changes in microbial community distribution and chemical composition during fermentation of Ampelopsis grossedentata leaves,fermented Ampelopsis grossedentata leaves were used as samples for microbial DNA extraction,PCR amplification,sequencing library construction,high-throughput sequencing data analysis,and identification of microbial community distribution. Based on the determination of the contents of flavonoids,polyphenols,proteins,amino acids,soluble sugars,and water-soluble extract,UPLC-MS/MS was applied to determine the changes in flavonoid and polyphenol compounds during fermentation. Comprehensive two-dimensional gas chromatography-time-of-flight mass spectrometry (GC×GC-TOFMS)system was usedto analyze the changes in volatile components before and after fermentation. The results showed that at the genus level,the dominant microbial communities in Ampelopsis grossedentata leaves were Lactobacillus,Pediococcus,and Bifidobacterium during fermentation. The contents of flavonoids,polyphenols,soluble sugars,and water-soluble extracts reached the maximum at 60 h and then decreased gradually during fermentation. Among the five flavonoid compounds (dihydromyricetin,myricitrin,piceatannol,myricetin,and quercetin),their contents had minor changes and strong stability. Among the eight polyphenol compounds (epigallocatechin gallate,gallocatechin,epigallocatechin,etc.),the content of epigallocatechin gallate (EGCG) decreased,while that epigallocatechin (EGC) increased,and the contents of the other polyphenol compounds remained basically unchanged. The contents of protein and amino acids decreased gradually throughout fermentation period. The main volatile components produced during fermentation included terpenes,alcohols,aldehydes,esters,and ketones,which contributed to the characteristic aromas of wood,floral,and mint notes;the key aroma components were methyl salicylate and caryophyllene.
