中文摘要: 为探究制备葵花籽蛋白抗氧化肽的最佳蛋白酶组合,并鉴定酶解物中的抗氧化肽段序列,利用不同双蛋白酶或三蛋白酶组合对葵花籽蛋白进行酶解,并通过测定不同酶解物的DPPH自由基清除率和总抗氧化能力以及对萎缩C2C12肌管中超氧化物歧化酶活性、丙二醛含量和谷胱甘肽含量的影响,进一步筛选最佳蛋白酶组合及酶解物中抗氧化能力最强的肽段分子量范围,通过肽段鉴定和抗氧化能力预测评分筛选出抗氧化肽段。结果表明,不同复合蛋白酶的酶解多肽中,胰蛋白酶、木瓜蛋白酶和碱性蛋白酶的复合酶解多肽抗氧化作用最强,其中分子量范围小于1 kDa的肽段抗氧化能力显著高于其他分子量肽段,并筛选出序列为LLPYYPN、LLPYYPNT、FSPHWTVN、GVAPGPVWTP和AIQSPHWT的抗氧化肽段。
Abstract:To investigate the optimal protease combination for the preparation of antioxidant peptides from sunflower seed protein and to identify antioxidant peptide sequences in the hydrolysates,sunflower seed protein was hydrolyzed using different combinations of dual or triple proteases. The antioxidant activities of the resulting hydrolysates were evaluated by measuring their DPPH free radical scavenging activity and total antioxidant capacity. In addition,their effects on superoxide dismutase activity,malondialdehyde content,and glutathione content in atrophic C2C12 myotubes were assessed to further screen the optimal protease combination and determine the peptide molecular weight fractions with the strongest antioxidant capacity. Antioxidant peptides were subsequently identified by peptide sequencing and antioxidant activity prediction scoring. The results showed that among the hydrolysates generated by different protease combinations,peptides produced by the combined hydrolysis with trypsin,papain,and alkaline protease exhibited the strongest antioxidant activity.Furthermore,peptides with a molecular weight below 1 kDa demonstrated significantly higher antioxidant capacity than those in other molecular weight fractions. Ultimately,five antioxidant peptides were identified,with the sequences LLPYYPN,LLPYYPNT,FSPHWTVN,GVAPGPVWTP,and AIQSPHWT.
