中文摘要: 为探讨食品天然活性成分羟基酪醇(hydroxytyrosol,HT)对动脉粥样硬化(atherosclerosis,AS)模型大鼠血脂代谢及血管内皮损伤的保护作用,以高脂饲料联合维生素D3和L-甲硫氨酸构建大鼠动脉粥样硬化模型,将大鼠随机分为正常组、模型对照组、HT低剂量组和HT高剂量组及阿托伐他汀组,连续灌胃干预后测定血清中血脂、肝功能及炎症因子等相关指标,采用苏木精-伊红染色、油红O染色观察肝脏与主动脉组织病理形态及脂质沉积情况。结果表明,该研究成功构建大鼠动脉粥样硬化模型;HT干预可显著调节血脂水平,改善肝功能指标,抑制氧化应激、降低血管损伤相关指标的活性,且高剂量效果更显著;病理切片显示,HT可明显减轻肝脏脂肪变性,减少肝脏及主动脉脂质沉积,缓解主动脉内膜损伤。综上,HT可通过调节血脂代谢、改善肝功能、抑制氧化应激与炎症反应、减轻血管内皮损伤及脂质沉积,对动脉粥样硬化模型大鼠发挥明显保护作用。
Abstract:To explore the protective effect of hydroxytyrosol (HT),a natural food active ingredient,on lipid metabolism and vascular endothelial injury in rats with atherosclerosis (AS),an AS rat model was established by feeding a high-fat diet combined with vitamin D3 and L-methionine. The rats were randomly divided into the normal group,model control group,HT low-dose group,HT high-dose group,and atorvastatin positive group.After continuous intragastric administration intervention,serum lipid profiles,liver function indexes,oxidative stress indicators and inflammatory factors were detected. Hematoxylin-eosin (HE) staining and Oil Red O staining were performed to observe the pathological morphology and lipid deposition of liver and aortic tissues.The results demonstrated that the rat AS model was successfully constructed. HT intervention significantly regulated serum lipid levels,improved liver function,inhibited oxidative stress,and reduced the activities of vascular injury-related indicators,with superior efficacy observed in the HT high-dose group. Pathological staining results further verified that HT markedly alleviated hepatic steatosis,reduced lipid deposition in the liver and aorta,and ameliorated aortic intimal injury. In conclusion,HT exerts prominent protective effects against atherosclerosis in model rats by regulating lipid metabolism,improving hepatic function,suppressing oxidative stress and inflammatory responses,and alleviating vascular endothelial damage and lipid accumulation. This study provides a reliable experimental basis for the further development of HT as a functional food factor and its application in dietary intervention for atherosclerosis.
