中文摘要: 为评价长双歧杆菌长亚种BL-01的益生特性及应用潜力，该研究从胃肠道耐受性、基因组特征及人体健康效应等方面对其进行系统分析。结果表明，长双歧杆菌长亚种BL-01在pH2.5人工胃液和pH8.0人工肠液中存活率分别为71.01%和85.75%，表现出良好的胃肠道耐受性。其基因组大小为2.60 Mbp，鸟嘌呤-胞嘧啶（guaninecytosine，GC）含量为60.31%，共含2 178 个编码序列（coding sequences，CDS）、61 个tRNA和8 个rRNA，并携带多种与环境耐受和潜在益生功能相关的基因。人群干预结果显示，BL-01可显著缓解便秘症状和旅行相关腹泻，显著增加双歧杆菌丰度（P＜0.01），降低大肠杆菌、肠球菌和粪球菌丰度（P＜0.05）；同时可促进儿童身高增长，提高骨钙素、抗酒石酸酸性磷酸酶（tartrate-resistant acid phosphatase，TRACP）水平，并改善骨面积和骨密度。综上，BL-01具有良好的耐受性、功能基因基础及健康促进作用。

Abstract:To systematically evaluate the probiotic properties and application potential of Bifidobacterium longum subsp. longum BL-01，this study investigated its gastrointestinal tolerance，genomic characteristics，and effects on human health. BL-01 demonstrated robust gastrointestinal tolerance，with survival rates of 71.01% in simulated gastric juice （pH2.5） and 85.75% in simulated intestinal juice （pH8.0）. Whole-genome draft analysis revealed that its genome comprised 2.60 Mbp with a guanine-cytosine （GC） content of 60.31%，encoding 2 178 coding sequences （CDS），61 tRNA genes，and 8 rRNA genes，and harbored multiple genes associated with environmental stress tolerance and putative probiotic functions. Human intervention demonstrated that BL-01 significantly alleviated constipation and travel-related diarrhea，markedly increased Bifidobacterium abundance （P＜0.01），and decreased Escherichia coli，Enterococcus，and fecal streptococci abundance （P＜0.05）.Furthermore，BL-01 promoted linear growth in children，as evidenced by elevated osteocalcin and tartrateresistant acid phosphatase （TRACP） levels，together with improved bone area and bone mineral density. In conclusion，BL-01 exhibits excellent gastrointestinal tolerance，a favorable functional genetic basis，and significant health-promoting effects.
