中文摘要: 该文以紫色红薯叶为原料,采用超声波辅助提取工艺提取其中的花色苷,并探究紫色红薯叶花色苷的抗氧化活性、抑制HCC细胞增殖及转移生物活性,采用pH示差法测定花色苷含量。通过单因素试验得到最佳提取工艺为柠檬酸浓度3%、提取时间35 min、提取温度45 ℃、提取功率350 W、料液比1∶30 (g/mL)。浓度为500 μg/mL的花色苷与维生素C对DPPH自由基清除能力均达到90%以上。当花色苷的浓度为140 μg/mL时,HCC细胞存活率仅为40.2%,证明紫色红薯叶花色苷有抑制HCC细胞存活的能力。划痕试验结果表明,紫色红薯叶花色苷对于HCC细胞的迁移有着明显的抑制作用。
Abstract:In this paper,by using purple sweet potato leaves as raw materials,anthocyanins were extracted by ultrasonic-assisted extraction technology. The antioxidant activity and biological activities related to the inhibition of HCC cell proliferation and metastasis of anthocyanins from purple sweet potato leaves were investigated,and the content of anthocyanins was determined by the pH differential method. The optimal extraction process was determined through single-factor experiments as follows:citric acid concentration of 3%,extraction time of 35 min,extraction temperature of 45 ℃,extraction power of 350 W,and material-to-liquid ratio of 1∶30 (g/mL).The DPPH radical scavenging rates of both anthocyanins and vitamin C at a concentration of 500 μg/mL exceeded 90%. When the anthocyanin concentration was 140 μg/mL,the survival rate of HCC cells was only 40.2%,indicating that anthocyanins from purple sweet potato leaves could inhibit the survival of HCC cells.Scratch test results indicated that anthocyanins from purple sweet potato leaves had a significant inhibitory effect on the migration of HCC cells.
