中文摘要: 为优化马鲛鱼副产物抗氧化肽的酶解工艺,并将其应用到鱼肠,该文以马鲛鱼碎肉为原料,通过单因素试验筛选最佳蛋白酶并优化其酶解条件;通过单因素和正交试验确定鱼肠的最佳配方。结果表明,最佳蛋白酶为中性蛋白酶,最佳酶解条件为中性蛋白酶添加量1 600 U/g、pH7、温度30 ℃,所得酶解液经10 kDa超滤纯化后,肽浓度从840.14 μg/mL提升至1 689.40 μg/mL,DPPH自由基清除率由44.71%提高至61.50%,还原力由0.38提升至0.55。通过正交试验确定马鲛鱼肠的最佳配方(以鱼糜质量为基准)为水添加量20%、玉米淀粉添加量40%、纯化肽粉添加量5.0%。与未添加抗氧化肽的对照组相比,添加抗氧化肽的马鲛鱼肠DPPH自由基清除率从35.23%显著提升至79.69%,还原力从0.29提升至0.63,且粗蛋白含量明显增加。
Abstract:The enzymatic hydrolysis process of antioxidant peptides from mackerel by-products was optimized and then the obtained antioxidant peptides were employed to prepare fish sausages. The minced mackerel meat was used as the raw material. Through single-factor experiments,the best protease was selected and its hydrolysis conditions were optimized. The formula of fish sausages was optimized through single-factor and orthogonal experiments. The results showed that the best protease was neutral protease,and the optimal hydrolysis conditions were neutral protease addition of 1 600 U/g,pH7,and 30 ℃. After purification with a 10 kDa ultrafiltration membrane,the peptide concentration of the hydrolysate increased from 840.14 μg/mL to 1 689.40 μg/mL.Moreover,the DPPH radical scavenging rate increased from 44.71% to 61.50%,and the reducing power increased from 0.38 to 0.55. Through orthogonal experiments,the formula of fish sausages (based on the mass of minced mackerel meat) was optimized as 20% water addition,40% corn starch addition,and 5.0% purified peptide powder addition. Compared with the control group without peptide addition,the addition of antioxidant peptides significantly increased the DPPH radical scavenging rate of mackerel sausages from 35.23% to 79.69% and the reducing power from 0.29 to 0.63. Furthermore,the content of crude protein also increased significantly.
