铁皮石斛多糖及联苄类化合物分步提取工艺优化

中文摘要: 为实现铁皮石斛多糖与联苄类化合物的分步高效提取,该研究采用单因素结合均匀设计法优化铁皮石斛多糖提取工艺;提取多糖后残渣直接用于铁皮石斛联苄类化合物的提取,在单因素试验基础上,采用响应面法优化联苄类化合物提取工艺。结果表明,多糖提取的最佳工艺条件为酶解温度65 ℃、酶解时间120 min、料液比1∶45 (g/mL)、加酶量1.5%,验证得到铁皮石斛多糖提取率为33.69%;联苄类化合物提取的最佳工艺条件为超声时间4 h、料液比1∶20(g/mL)、乙醇体积分数70%,验证得到铁皮石斛联苄类化合物提取率平均值为3.71%。预测值与试验实际值相近,说明试验结果可靠,优化的提取工艺可行有效。
Abstract:To realize the efficient stepwise extraction of Dendrobium officinale polysaccharides and bibenzyl compounds,single-factor experiments combined with uniform design were employed in this study to optimize the extraction process of D.officinale polysaccharides. The residue obtained after polysaccharide extraction was directly reused for the extraction of bibenzyl compounds from D. officinale. On the basis of single-factor tests,response surface methodology was further applied to optimize the extraction technology of bibenzyl compounds.The results showed that the optimal conditions for polysaccharide extraction were enzymatic hydrolysis temperature of 65 ℃,enzymatic hydrolysis time of 120 min,solid-liquid ratio of 1∶45 (g/mL),and enzyme dosage of 1.5%. Under the verified conditions,the extraction rate of Dendrobium officinale polysaccharides reached 33.69%. The optimal conditions for the extraction of bibenzyl compounds were ultrasonic extraction time of 4 h,solid-liquid ratio of 1∶20 (g/mL),and ethanol volume fraction of 70%. Under verification experiments,the average extraction rate of bibenzyl compounds from Dendrobium officinale was 3.71%. The predicted values were in good agreement with the experimental measured values,which indicated that the experimental results were reliable and the optimized extraction process was feasible and effective.
