中文摘要: 肠炎是导致老年人群营养失衡和健康恶化的主要疾病之一。该文基于在线数据库、网络药理学和分子对接技术,探究来自海洋紫球藻苏氨酰酪氨酸(threonine-tyrosine,TY)肽对老年性肠炎与衰老的潜在共调节作用机制。采用在线数据库获取肠炎、衰老和TY肽的相关靶点,构建蛋白-蛋白互作网络并筛选核心靶点,利用分子对接验证核心靶点和TY肽的结合亲和力,并对核心靶点进行共表达和功能分析及构建转录因子(transcription factor,TF)-miRNA-mRNA调控网络。结果显示,韦恩图共筛选出54 个交集靶点。根据拓扑值分析筛选出14 个核心靶点。对作为炎症调节的重要靶点即排名前5 个核心靶点进行分子对接和功能分析可知,TY肽的每个对接模型均与前5 个核心靶点相互作用,并表现出较好的结合亲和力。此外,TF-miRNA-mRNA调控网络分析预测hsa-miR-216a-3p和hsa-miR-6726-5p共同调控排名前5 的核心靶点。通过初步揭示TY肽在改善肠炎和延缓衰老方面的靶点与机制,为开发针对老年人的TY肽保健食品提供了理论依据。
Abstract:Enteritis is one of the major diseases leading to nutritional imbalances and health deterioration in the elderly population. Based on online databases,network pharmacology,and molecular docking techniques,this study investigated the potential co-regulatory mechanisms of threonine-tyrosine (TY),a peptide derived from the marine alga Porphyridium cruentum,in managing age-related enteritis and senescence. Relevant targets associated with enteritis,aging,and TY peptide were retrieved from online databases. A protein-protein interaction (PPI) network was constructed to screen core targets. Molecular docking was performed to validate the binding affinity between the core targets and TY peptide. Subsequently,co-expression and functional analyses of the core targets were conducted,and a transcription factor (TF)-miRNA-mRNA regulatory network was established. A Venn diagram analysis identified 54 overlapping targets. Based on topological parameters,14 core targets were further screened. Molecular docking and functional analyses of the top five core targets,which play key roles in inflammatory regulation,revealed that each docking model of TY peptide interacted with these five core targets and exhibited favorable binding affinities. Furthermore,analysis of the TF-miRNA-mRNA regulatory network predicted that hsa-miR-216a-3p and hsa-miR-6726-5p co-regulate the top five core targets.This study preliminarily elucidates the targets and mechanisms by which TY peptide may ameliorate enteritis and delay aging,providing a theoretical basis for the development of TY peptide-based functional foods tailored for the elderly population.
