中文摘要: 为获得可以生产舍雷肽酶的粘质沙雷氏菌(Serratia marcescens),对一株通过添加酪氨酸为底物的筛选培养基筛选获得的产红色色素的菌株进行形态学、生理生化及分子生物学等鉴定,确定该菌株为粘质沙雷氏菌,并将其命名为Cczu.wj01。在初筛液体培养基中进行培养产酶,发现该菌株酶活为23.44 U/L。为进一步挖掘该菌株产酶潜力,对其发酵条件进行优化,确定Cczu.wj01的最优发酵培养基组分为麦芽浸膏5 g/L、大豆蛋白胨3 g/L、氯化钠10g/L、磷酸氢二钾10mmol/L。最优发酵条件为初始pH值6.5、种龄12 h、接种量0.2%、温度31 ℃、摇床转速160 r/min,在最优发酵条件下培养16h后,菌株Cczu.wj01所产舍雷肽酶酶活达到316.42U/L。
Abstract:To obtain Serratia marcescens capable of producing serratiopeptidase,a red-pigment-producing strain isolated using a tyrosine-supplemented screening medium was subjected to morphological,physiologicalbiochemical,and molecular biological identification. The strain was identified as Serratia marcescens and designated as S. marcescens Cczu.wj01. When cultured in the initial screening liquid medium,the strain exhibited an enzyme activity of 23.44 U/L. To further explore its enzyme-producing potential,the fermentation conditions were optimized. The optimal medium composition for Cczu. wj01 was determined as follows:malt extract 5 g/L,soy peptone 3 g/L,sodium chloride 10 g/L,and dipotassium hydrogen phosphate 10 mmol/L. The optimal fermentation conditions were an initial pH of6.5,a seed age of 12 h,an inoculum size of0.2%,a culture temperature of 31 ℃,and a shaking speed of 160 r/min. After 16 h of cultivation under the optimized conditions,the serratiopeptidase activity of strain Cczu.wj01 reached 316.42U/L.
