中文摘要: 为实现刺梨汁加工副产物高值化利用,采用复合微生物发酵-酶解耦联技术制备刺梨汁加工副产物活性肽,并开发刺梨汁加工副产物多肽口服液。该文通过单因素与正交试验优化发酵、蛋白提取、酶解及口服液配方。结果表明:4株复合益生菌(鼠李糖乳杆菌、嗜酸乳杆菌、青春双歧杆菌、植物乳植杆菌1∶1∶1∶1,体积比)在36 ℃发酵20d最优,刺梨汁加工副产物粗蛋白含量由10.61 g/100g提升至13.54 g/100g(提升27.6%),总糖含量、单宁含量等抗营养因子明显降低;采用碱溶酸沉法(pH10、51 ℃超声49 min)提取刺梨汁加工副产物分离蛋白,蛋白提取率达17.35%;最优酶解条件为菠萝蛋白酶∶中性蛋白酶1∶1(体积比)、酶解温度55 ℃、酶解pH值7.5、酶解时间1.0 h,此条件下肽得率为29.45%,较单一菠萝蛋白酶提升48.06%。口服液最优配方为阿拉伯糖添加量10%、罗汉果甜苷添加量0.10%、刺梨多肽添加量5%,稳定剂为0.10%羧甲基纤维素钠(carboxymethylcellulose sodium,CMC-Na),此条件下感官评分为85.2、离心沉淀率为3.92%。
Abstract:To realize the high-value utilization of Rosa roxburghii juice processing by-products,a coupled technology of compound microbial fermentation and enzymatic hydrolysis was adopted to prepare bioactive peptides from these by-product,and a novel Rosa roxburghii polypeptide oral liquid was further developed. In this study,single-factor and orthogonal experiments were performed to optimize the key parameters of fermentation,protein extraction,enzymatic hydrolysis,and oral liquid formulation. The results indicated that the optimal fermentation conditions were established using a four-strain compound probiotic system (Lacticaseibacillus rhamnosus,Lactobacillus acidophilus,Bifidobacterium adolescentis,and Lactiplantibacillus plantarum with an equal volume ratio of 1∶1∶1∶1,a fermentation temperature of36 ℃,and a fermentation duration of20d. After fermentation,the crude protein content of Rosa roxburghii juice processing by-product increased from 10.61 g/100 g to 13.54 g/100 g,representing a 27.6% elevation,while the contents of anti-nutritional factors including total sugar and tannin were markedly reduced. Protein isolates were extracted from the by-products via the alkali solution and acid precipitation method with ultrasonic assisted under the conditions of pH10,51 ℃,and 49 min of ultrasonic treatment,achieving a protein extraction rate of 17.35%. The optimal enzymatic hydrolysis conditions were as follows:bromelain to neutral protease=1∶1(volume ratio),hydrolysis temperature of 55 ℃,hydrolysis pH value of 7.5,and hydrolysis time of 1.0 h. Under these optimal conditions,the peptide yield reached 29.45%,which was 48.06% higher than that of the single bromelain hydrolysis group. The optimal formulation of the polypeptide oral liquid was determined as 10% arabinose,0.10% mogroside,5% Rosa roxburghii polypeptide,and 0.10% carboxymethylcellulose sodium (CMC-Na) as the stabilizer. The finished oral liquid prepared with this optimal formula achieved a sensory evaluation score of 85.2 and a centrifugal precipitation rate of3.92%.
