超声辅助酶法提取菊花多糖工艺优化及其抗氧化作用

中文摘要: 该研究在单因素试验的基础上,结合响应面优化试验,以多糖得率为指标,优选超声辅助酶法提取菊花多糖的最佳工艺;采用高效液相色谱(high performance liquid chromatography,HPLC)-衍生化法测定菊花多糖的单糖组成并测定其抗氧化能力。结果表明,菊花多糖最佳的提取条件为提取时间99 min、提取温度52 ℃、料液比1∶36.5(g/mL)、酶添加量460mg,在此条件下菊花多糖得率为(5.84±0.32)%。对菊花多糖的单糖组成进行分析,发现菊花多糖由甘露糖、葡萄糖醛酸、葡萄糖和岩藻糖组成,物质的量比为1.00∶5.44∶1.55∶16.90。体外抗氧化试验结果表明,浓度为5.0 mg/mL时,菊花多糖的ABTS+自由基清除率为(51.12±6.38)%、DPPH自由基清除率为(56.39±2.76)%。该研究优化得到的菊花多糖超声辅助酶法提取工艺优于其他提取工艺(热水提取、超声辅助提取和酶法提取),菊花多糖提取率显著增加,为进一步将菊花多糖开发成抗氧化剂、保健食品等提供参考。
Abstract:On the basis of single-factor experiments,this study combined response surface optimization tests with polysaccharide yield as the evaluation index to screen the optimal process for ultrasound-assisted enzymatic extraction of chrysanthemum polysaccharides. High performance liquid chromatography (HPLC) combined with derivatization method was used to determine the monosaccharide composition and antioxidant capacity of chrysanthemum polysaccharides. The results showed that the optimal extraction conditions were extraction time of99 min,extraction temperature of52 ℃,solid-liquid ratio of 1∶36.5(g/mL) and enzyme dosage of 460 mg. Under these conditions,the yield of chrysanthemum polysaccharides reached (5.84±0.32)%.Monosaccharide composition analysis revealed that chrysanthemum polysaccharides consisted of mannose,glucuronic acid,glucose and fucose at a molar ratio of 1.00∶5.44∶1.55∶16.90. In-vitro antioxidant tests indicated that at the concentration of 5.0 mg/mL,the ABTS+radical scavenging rate of chrysanthemum polysaccharides was (51.12±6.38)%,and the DPPH radical scavenging rate was (56.39±2.76)%. The optimized ultrasound-assisted enzymatic extraction process was superior to hot-water extraction,single ultrasound extraction and single enzymatic extraction,which significantly improved the extraction yield of chrysanthemum polysaccharides. This study provides a reference for further development of chrysanthemum polysaccharides as natural antioxidants and functional foods.
