复合酶法提取西州蜜瓜皮多糖工艺优化及其抗氧化活性

中文摘要: 以粗多糖提取率为主要指标,系统优化西州蜜瓜皮多糖复合酶法提取工艺。通过单因素试验和正交试验确定最优提取条件,并对所得粗多糖进行脱色、除蛋白纯化处理,分别得到脱色多糖和脱色脱蛋白多糖。同时,进一步分析多糖的粒径分布、Zeta电位变化及其对DPPH自由基与ABTS+自由基的清除能力。结果表明:西州蜜瓜皮多糖最佳提取工艺为复合酶质量比1∶2、酶用量2.8%、酶解时间16min、pH6.5、酶解温度40 ℃,该条件下粗多糖提取率达(12.40±0.10)%。脱色脱蛋白多糖的平均粒径由421.7nm下降至310.8 nm,Zeta电位从-17.54mV升高至-13.77 mV。抗氧化活性分析显示,粗多糖对DPPH和ABTS+自由基的清除率分别为59.52%和61.71%,均高于脱色多糖和脱色脱蛋白多糖。综上,西州蜜瓜皮多糖具有较好的体外抗氧化活性。
Abstract:This study aimed to systematically optimize the compound enzymatic extraction process of polysaccharides from the green peel of Xizhou cantaloupe,using the crude polysaccharide extraction yield as the primary index. Through single-factor experiments and an orthogonal array test,the optimal extraction conditions were determined. The obtained crude polysaccharides were purified by decolorization and deproteinization to obtain decolorized polysaccharides and decolorized-deproteinized polysaccharides,respectively. Furthermore,the particle size distribution,Zeta potential changes,and DPPH and ABTS+radical scavenging capacities of the polysaccharides were analyzed. The results indicated that the optimal extraction conditions were as follows:enzyme mass ratio 1∶2,enzyme dosage of 2.8%,hydrolysis time of 16 min,pH6.5,and hydrolysis temperature 40 ℃. Under these conditions,the polysaccharide extraction yield reached (12.40±0.10)%. For the decolorized-deproteinized polysaccharides,the average particle size decreased from 421.7 nm to 310.8 nm,while the Zeta potential increased from -17.54 mV to -13.77 mV. Antioxidant activity analysis showed that the scavenging rates of the crude polysaccharides against DPPH and ABTS+free radicals were 59.52% and 61.71%,respectively,both higher than those of the decolorized and the decolorized-deproteinized polysaccharides. In conclusion,the green peel polysaccharides of Xizhou cantaloupe exhibit promising in vitro antioxidant activity.
