思茅甜笋粗多糖的提取及其抗氧化活性

中文摘要: 为优化思茅甜笋多糖提取工艺,研究其抗氧化能力,采用内部沸腾法对思茅甜笋多糖进行提取,并对提取条件进行单因素试验优化,对其单糖组成和抗氧化活性进行测定。单因素试验得出的最佳条件为解析温度60 ℃、提取时间6 min、解析时间15 min、料液比1∶30(g/mL)、解析剂用量6 mL、解析剂浓度30%,此条件下多糖的平均得率为7.5%。通过单糖组成和分子量分析,该工艺条件下获得的多糖主要由鼠李糖、葡萄糖、半乳糖、木糖和甘露糖组成,重均分子量为100 000 Da。通过抗氧化活性测试,得出质量浓度为5.0 mg/mL的思茅甜笋粗多糖对羟自由基和超氧阴离子自由基的清除率分别可达1.0 mg/mL L-抗坏血酸的52.18%、81.33%,表明其对两种自由基均具有一定的清除能力,且对超氧阴离子自由基的清除效果更好;1.0 mg/mL的思茅甜笋粗多糖对1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picryl-hydrazyl radical,DPPH)自由基的清除能力相当于同浓度L-抗坏血酸的50.58%;总还原能力测定结果也表明,思茅甜笋多糖具有较好的还原能力。
Abstract:To optimize the extraction process and evaluate the antioxidant activity of polysaccharides from Simao sweet bamboo shoots,the internal boiling method was employed. Single-factor experiments were conducted to optimize the extraction conditions,and the monosaccharide composition as well as antioxidant activity of the obtained polysaccharides were determined. The optimal conditions were as follows:resolution temperature 60 ℃,extraction time 6 min,resolution time 15 min,solid-to-liquid ratio 1∶30 (g/mL),resolving agent volume 6 mL,and resolving agent concentration 30%. Under these conditions,the average yield of polysaccharides was 7.5%. Monosaccharide composition and molecular weight analysis revealed that the polysaccharides were mainly composed of rhamnose,glucose,galactose,xylose,and mannose,with a weight-average molecular weight of 100 000 Da. Antioxidant activity assays showed that at a concentration of 5.0 mg/mL,the polysaccharides from Simao sweet bamboo shoots exhibited hydroxyl radical and superoxide anion radical scavenging activities equivalent to 52.18% and 81.33%,respectively,of that of 1.0 mg/mL L-ascorbic acid,indicating that the polysaccharides possess notable radical scavenging ability,with better efficacy against superoxide anion radicals. At 1.0 mg/mL,the crude polysaccharides showed DPPH radical scavenging activity equivalent to 50.58% of that of L-ascorbic acid at the same concentration. The total reducing power assay further confirmed that the polysaccharides from Simao sweet bamboo shoots have good reducing capacity.
