夏秋茶多糖复合酶辅助超声提取工艺、结构表征及抗氧化活性

中文摘要: 为优化夏秋茶多糖提取工艺、解析其结构特征并评价其抗氧化活性,该文以信阳夏秋茶为原料,采用复合酶辅助超声提取法,基于单因素、Box-Behnken响应面法对夏秋茶多糖提取工艺进行优化;以高效凝胶渗透色谱、红外光谱、热重分析及X-射线衍射分析对其结构进行表征;通过DPPH·、ABTS+·清除试验评价其体外抗氧化能力。结果表明:最佳工艺条件为复合酶(纤维素酶∶果胶酶∶中性蛋白酶=1∶1∶1,质量比)添加量12%、酶解温度45 ℃、酶解时间60 min、超声温度60 ℃、超声时间30 min、超声功率280 W,此工艺条件下夏秋茶多糖提取率达12.3%。夏秋茶多糖为酸性杂多糖,具有疏松孔状结构,由表面光滑的不规则片状结构聚集而成,内部同时存在晶体与无定形结构,在163.9 ℃以下具有较好的热稳定性。抗氧化活性试验表明,夏秋茶多糖对DPPH·、ABTS+·的半抑制浓度(half maximal inhibitory concentration,IC50)值分别为0.39、0.57 mg/mL,说明其具备较好的抗氧化活性。
Abstract:To optimize the extraction process of summer and autumn tea polysaccharides,analyze their structural characteristics,and evaluate their antioxidant activity,Xinyang summer-autumn tea was used as the raw material,polysaccharides were extracted by compound enzyme-assisted ultrasonic extraction. The extraction process was optimized based on single-factor experiments and Box-Behnken response surface methodology.The structure was characterized by high-performance gel permeation chromatography,infrared spectroscopy,thermogravimetric analysis,and X-ray diffraction. The in vitro antioxidant activity was evaluated by DPPH radical and ABTS+ radical scavenging assays. The results showed that the optimal process conditions were as follows:compound enzyme (cellulase∶pectinase∶neutral protease=1∶1∶1,mass ratio) dosage of 12%,enzymatic hydrolysis temperature of 45 ℃,enzymatic hydrolysis time of 60 min,ultrasonic temperature of 60 ℃,ultrasonic time of 30 min,and ultrasonic power of 280 W. Under these conditions,the extraction yield of polysaccharides from summer and autumn tea reached 12.3%. The polysaccharides from summer and autumn tea were acidic heteropolysaccharides with a loose and porous structure,which were formed by the aggregation of irregular flake structures with smooth surfaces. Both crystalline and amorphous structures existed inside,and the polysaccharides possessed good thermal stability below 163.9 ℃. The antioxidant activity tests showed that the half maximal inhibitory concentrations (IC50)value of polysaccharides against DPPH· and ABTS+· were 0.39 mg/mL and 0.57 mg/mL,respectively,indicating that the polysaccharides had favorable antioxidant activity.
