超声波辅助提取枸杞子粗多糖及其功能性评价

中文摘要: 该文以提取液中枸杞子粗多糖含量为指标,采用单因素试验考察超声时间、超声温度、超声功率等对提取液粗多糖含量的影响,在此基础上,通过响应曲面法优化枸杞子粗多糖的提取条件。结果表明,最佳的提取工艺为超声时间57.3 min、超声温度86.5 ℃、超声功率164 W,在此条件下,提取液粗多糖的含量为(0.631 6±0.002 6) mg/mL。利用吸附、沉淀分离等方法去除提取液中的色素、黄酮等成分,进一步获得了粗多糖纯化样品,浓度为2.5 mg/mL的粗多糖样品对DPPH自由基清除能力达到80%以上。细胞实验结果表明,随着枸杞子粗多糖浓度的增加,肝细胞癌(hepatocellular carcinoma,HCC)细胞的存活率逐渐降低,当枸杞子粗多糖的浓度达到140 μg/mL时,存活率仅为44.4%,证明枸杞子粗多糖有抑制HCC细胞存活的能力。通过划痕实验证明,枸杞子粗多糖提取液对于HCC细胞具有良好的抑制转移能力。
Abstract:Taking Lycium barbarum crude polysaccharide content in the extract as the index,a single factor test was conducted to investigate the effects of ultrasonic time,temperature,and power on the crude polysaccharide content in the extract. Then,response surface experiment was performed to optimize the extraction conditions of Lycium barbarum crude polysaccharides. According to the results,the optimum ultrasonic extraction process was a time of 57.3 min,a temperature of 86.5 ℃,and a power of 164 W. Under these conditions,crude polysaccharide content in the extract was (0.631 6±0.002 6) mg/mL. Pigments and flavonoids in the extract were separated by adsorption and precipitation to obtain purified crude polysaccharide samples. At a concentration of 2.5 mg/mL,the DPPH free radical scavenging rate of the crude polysaccharide sample reached 80%.The cell experiment results showed that as the concentration of Lycium barbarum crude polysaccharides was increased,the survival rate of hepatocellular carcinoma (HCC) cells was gradually decreased. When the concentration of Lycium barbarum crude polysaccharides reached 140 μg/mL,the survival rate of HCC cells was only 44.4%,proving that the crude polysaccharides could inhibit HCC cell survival. Scratch experiments demonstrated that Lycium barbarum crude polysaccharide extract exhibited a good inhibitory effect on HCC cell metastasis.
