中文摘要: 采用复合酶(纤维素酶和果胶酶)协同超声辅助乙醇法提取桑枝皮中1-脱氧野尻霉素(1-deoxynojirimycin,DNJ),研究纤维素酶与果胶酶质量比、复合酶质量浓度、乙醇体积分数、pH值、超声时间、超声温度对桑枝皮DNJ提取率的影响,进一步选择复合酶质量浓度、超声时间和乙醇体积分数作为变量,采用响应面法优化提取条件,得到最佳工艺条件为纤维素酶和果胶酶质量比1∶1、复合酶质量浓度4.82 mg/mL、乙醇体积分数64.6%、pH3.0、超声时间17.8 min、超声温度30 ℃,在该条件下桑枝皮的DNJ提取率理论值为0.845%,实际提取率为0.820%。
Abstract:This study aimed to optimize the extraction of 1-deoxynojirimycin (DNJ) from mulberry branch bark using a combined enzymatic (cellulase and pectinase) and ultrasound-assisted ethanol method. The effects of the cellulase-to-pectinase mass ratio,enzyme complex mass concentration,ethanol volume fraction,pH value,ultrasound time,and temperature on DNJ extraction efficiency were systematically investigated. Enzyme concentration,ultrasound time,and ethanol volume fraction were further optimized using Box-Behnken response surface methodology. The optimal extraction conditions were identified as a cellulase-to-pectinase mass ratio of 1∶1,enzyme complex mass concentration of 4.82 mg/mL,ethanol volume fraction of 64.6%,pH3.0,ultrasound time of 17.8 min,and ultrasound temperature of 30 ℃. Under these conditions,the theoretical DNJ extraction yield from mulberry branch bark was 0.845%,and the actual extraction yield was 0.820%.
