超声辅助碱法提取菰米蛋白工艺优化及其功能性质评价

中文摘要: 为提高菰米蛋白提取率并改善其功能性质,该研究采用超声辅助碱法提取菰米蛋白,利用响应面法优化提取工艺,并与传统碱法提取效果进行对比;通过测定持水性、持油性、乳化性、起泡性等功能特性,结合平均粒径、Zeta电位及红外光谱分析,研究超声处理对菰米蛋白结构及功能的影响。结果表明,最优提取工艺为液料比28∶1 (mL/g)、提取时间30 min、pH12、提取温度38 ℃,此时蛋白提取率达87.81%。超声处理使菰米蛋白平均粒径减小、Zeta电位绝对值增大,且二级结构中α-螺旋与β-折叠比例降低,β-转角与无规卷曲比例上升,表明其结构更为松散舒展。超声辅助碱法提取得到的菰米蛋白的持水性、持油性、乳化性、乳化稳定性及起泡稳定性均显著优于传统碱法(P<0.05)。
Abstract:To improve the extraction efficiency of wild rice protein and enhance its functional properties, ultrasound-assisted alkaline extraction was employed to extract wild rice protein. The extraction process was optimized using response surface methodology and compared with the traditional alkaline extraction method. By determining functional properties including water-holding capacity, oil-holding capacity, emulsifying properties, and foaming properties, combined with particle size, Zeta potential, and infrared spectroscopy analysis,the effects of ultrasound treatment on the structure and function of wild rice protein were investigated. The results showed that the optimal extraction conditions were as follows: liquid-to-solid ratio of 28∶1 (mL/g), extraction time of 30 min, pH12, and extraction temperature of 38 ℃, under which the protein extraction rate reached 87.81%. Ultrasound treatment reduced the particle size of wild rice protein and increased the absolute value of the Zeta potential. Meanwhile, the contents of α-helix and β-sheet in the secondary structure decreased, whereas the proportions of β-turn and random coil increased, indicating a looser and more extended structure. The water-holding capacity, oil-holding capacity, emulsifying property, emulsion stability, and foaming stability of wild rice protein extracted by the ultrasound-assisted alkaline method were all significantly higher than those obtained by the traditional alkaline method (P<0.05).
