低共熔溶剂提取法对大球盖菇多糖理化性质的影响

中文摘要: 为探究低共熔溶剂(deep eutectic solvents,DES)提取法对大球盖菇多糖的影响,以大球盖菇为原料,比较低共熔溶剂提取法和水提法提取大球盖菇多糖理化性质的差异。结果表明,与水提法相比,采用低共熔溶剂提取大球盖菇多糖的提取率提高67.07%,纯度无显著差异。傅里叶变换红外光谱结果显示,两种提取方法得到的多糖均具有典型的多糖特征吸收峰且为酸性多糖。扫描电子显微镜结果显示,两种多糖表面均呈鳞片状,但片状形态和规则差异较大。单糖组成结果显示,两种提取方法得到的多糖均由10种相同的单糖组成,但低共熔溶剂提取法得到大球盖菇多糖的糖醛酸相对含量较高。差示扫描量热(differential scanning calorimeter,DSC)仪分析结果显示,低共熔溶剂提取的多糖吸热焓值较水提法提高16%。低共熔溶剂提取法得到的大球盖菇多糖乳化活性指数和乳化稳定性指数均高于水提法。抗氧化活性结果表明,低共熔溶剂提取法得到的大球盖菇多糖抗氧化活性明显高于水提法。
Abstract:To investigate the effect of deep eutectic solvent (DES) extraction on the polysaccharides of Stropharia rugosoannulata, this study used Stropharia rugosoannulata as the raw materials and compared the differences in physicochemical properties of the polysaccharides obtained by DES extraction and traditional water extraction. The results showed that compared with water extraction, the extraction rate of Stropharia rugosoannulata polysaccharides by DES extraction was increased by 67.07%, with no significant difference in purity.Fourier transform infrared spectroscopy analysis indicated that the polysaccharides obtained by both extraction methods had typical characteristic absorption peaks of polysaccharides and were identified as acidic polysaccharides. Scanning electron microscope (SEM) observations showed that the surfaces of both polysaccharide samples were scaly, but significant differences were observed in their flake morphology and regularity. Monosaccharide composition analysis revealed that the polysaccharides from both extraction methods consisted of the same 10 monosaccharides. However, the relative content of uronic acid in the DES-extracted polysaccharides was higher. DSC analysis demonstrated that the endothermic enthalpy value of the DES-extracted polysaccharides was 16% higher than that of the water-extracted ones. Furthermore, both the emulsifying activity index (EAI) and emulsifying stability index (ESI) of the DES-extracted polysaccharides were higher than those of the water-extracted polysaccharides. Antioxidant activity evaluation showed that the antioxidant capacity of the Stropharia rugosoannulata polysaccharides extracted by DES was significantly superior to that extracted by water extraction.
