Abstract:Pediococcus spp. as an emerging lactic acid bacterium has attracted great attention due to its fermentation ability and probiotic properties. At present，the quantitative detection method of Pediococcus has not been stipulated in the national standard，and there is no specific isolation medium for Pediococcus. To determine the most suitable isolation medium and related conditions，this study compared the ability of 3 media to isolate Pediococcus. Pediococcus pentosaceus，Pediococcus acidilactici，Lactobacillus plantarum，and Enterococcus faecalis were inoculated into triphenyl tetrazolium chloride （TTC）+MRS agar medium，AMRS agar medium，and modified MRS agar medium to study the screening characteristics of different culture media. The selective counting ability of the culture media could be evaluated，and the quantitative detection method of Pediococcus could be established. Studies had shown that the selective counting ability of the cysteine hydrochloride，vancomycin，and novobiocin modified MRS agar media was best and could be used for the quantitative detection of the Pediococcus.
