双响应值联合优化鲽鱼骨汤工艺及其关键呈味特性解析

中文摘要: 以鲽鱼骨为研究对象,在单因素试验基础上,以整体接受度和可溶性蛋白含量为指标,对鲽鱼骨汤制备工艺进行响应面优化试验,并采用液相色谱-串联质谱对鲽鱼骨汤中游离氨基酸和呈味核苷酸进行靶向代谢组学分析。结果表明,鲽鱼骨汤的最佳工艺条件为烤制时间6 min、熬煮时间70 min、腌料添加量6%。在此条件下熬制出的鲽鱼骨汤汤汁乳白、风味浓郁,整体接受度为9.2,可溶性蛋白含量的测定结果为49.02 mg/mL,该结果与模型的预测值之间无显著差异。经工艺优化后的鲽鱼骨汤,游离氨基酸总量达1 196.78 μg/mL,与对照组相比提高了140%,其中鲜味氨基酸和甜味氨基酸含量明显增加,呈味核苷酸总量提高至56.34 μg/mL。
Abstract:This study focuses on optimizing the preparation process of Pleuronectes platessa bone soup. Based on single-factor experiments,response surface methodology was employed using overall acceptability and soluble protein content as evaluation criteria. Targeted metabolomic analysis of free amino acids and flavor nucleotides in Pleuronectes platessa bone soup was performed using liquid chromatography-tandem mass spectrometry. The results demonstrated that the optimal process conditions were as follows:roasting time of 6 min,simmering time of 70 min,and seasoning addition of 6%. Under these conditions,the prepared bone soup appeared milky white in color and exhibited a rich flavor,achieving an overall acceptability score of 9.2. The measured soluble protein content was 49.02 mg/mL,and there was no significant difference between this result and the predicted value from the model. After process optimization,the total free amino acid content in the soup increased to 1 196.78 μg/mL,representing a 140% improvement compared with the control group. The optimized process led to a significant increase in umami and sweet amino acids,along with an increase in the total flavor nucleotide content to 56.34 μg/mL.
