中文摘要: 为探究红枣在生产过程中产生的废渣中色素的关键浸提工艺,并分析该色素的抗氧化活性和挥发性成分组成。以色价值为指标,在单因素试验基础上,采用响应面分析法优化红枣色素关键浸提参数,并对该色素浸提物进行抗氧化活性检测及挥发性成分鉴定。结果显示,红枣色素的最佳浸提参数为浸提温度60 ℃、浸提时间10 h、KOH浓度0.2 mol/L,在此条件下经过三级色素浸提试验验证,每级色价值分别为0.598、0.625、0.506,相对误差分别为8.79%、4.73%、22.92%。抗氧化活性测定结果表明,该色素具有清除DPPH自由基、ABTS+自由基和羟基自由基的作用,其自由基清除效果与色素溶液浓度呈正相关,半数抑制浓度(half maximal inhibitory concentration,IC50)值分别为0.145 2、0.289 2、0.449 3 mg/mL。顶空固相微萃取-气相色谱-质谱联用技术分析鉴定出匹配度大于50的红枣色素中挥发性成分共有23 种,包含酮类12 种、醇类2 种、醛类2 种、酯类4 种、烯类1种及其他类2 种,其中酮类占比最大,相对含量为74.27%。
Abstract:This study aimed to optimize the key extraction parameters of pigment from jujube residues generated during jujube processing,and to analyze the antioxidant activity and volatile components of the extracted pigment. Using color value as the response indicator,single-factor tests and response surface methodology were employed to optimize the extraction process. The optimized pigment extract was then subjected to antioxidant activity assays and volatile component analysis. The results showed that the optimal extraction conditions were an extraction temperature of 60 ℃,an extraction time of 10 h,and a KOH concentration of 0.2 mol/L. Under these conditions,a three-stage extraction process yielded color values of 0.598,0.625,and 0.506 for each stage,with relative errors of 8.79%,4.73%,and 22.92%,respectively. Antioxidant activity assays demonstrated that the pigment possessed DPPH free radical,ABTS+ free radical,and hydroxyl free radical scavenging activities in a concentration-dependent manner,with half maximal inhibitory concentration values of 0.145 2,0.289 2,and 0.449 3 mg/mL. Headspace solid-phase microextraction-gas chromatography-mass spectrometry(HS-SPME-GC-MS) analysis identified 23 volatile components in the jujube pigment with a match degree greater than 50,including 12 ketones (74.27% relative content),2 alcohols,2 aldehydes,4 esters,1 alkenes,and 2 other compounds,with ketones being the predominant class.
