中文摘要: 为提高库页红景天中红景天苷、多酚等活性成分的提取效率及其抗氧化活性,该研究采用超声-无酶、超声-单一酶(果胶酶或纤维素酶)以及超声-复合酶(果胶酶+纤维素酶)3 种处理方式,对比不同酶解处理对库页红景天活性成分的提取效果及其抗氧化活性的影响。采用高效液相色谱法与福林酚比色法分别测定提取物中红景天苷含量与多酚提取率,通过DPPH自由基清除试验、ABTS+自由基清除试验和总还原能力试验评价提取物的抗氧化活性。结果表明,超声-复合酶法可显著提高红景天苷含量和多酚的提取率,分别达15.53 mg/g和13.31%;库页红景天提取物在DPPH自由基清除试验和ABTS+自由基清除试验中表现出极高活性,清除率分别为87.9%和99.8%,且在总还原能力试验中表现较佳。研究结果显示,超声-复合酶处理可有效提升库页红景天中活性成分的提取效率和提取物抗氧化性能,其中酶的协同作用发挥关键作用。
Abstract:To improve the extraction efficiency of active components such as salidroside and polyphenols from Rhodiola saccharinensis Boriss,as well as their antioxidant activity,three treatment methods were compared in this study:ultrasound-assisted enzyme-free extraction,ultrasound-assisted single-enzyme extraction (pectinase or cellulase),and ultrasound-assisted composite-enzyme extraction (pectinase+cellulase). The effects of different enzymatic treatments on the extraction efficiency and antioxidant activity of Rhodiola saccharinensis Boriss were investigated. The salidroside content and polyphenol extraction rate in the extracts were determined by high-performance liquid chromatography (HPLC) and the Folin-Ciocalteu colorimetric method,respectively.Meanwhile,the antioxidant activity of the extracts was evaluated using DPPH and ABTS+ free radical scavenging assays,as well as a total reducing power test. The results showed that the ultrasound-assisted compositeenzyme treatment significantly increased the salidroside content and polyphenol extraction rate,reaching 15.53 mg/g and 13.31%,respectively. The extracts exhibited excellent free radical scavenging activity in the DPPH and ABTS+ assays,with scavenging rates of 87.9% and 99.8%,respectively,and also performed well in the total reducing power test. These findings indicated that ultrasound-assisted composite-enzyme treatment could effectively enhance the extraction efficiency of active components and the antioxidant performance of Rhodiola saccharinensis Boriss,in which the synergistic effect of enzymes played a crucial role.
