中文摘要: 为实现密蒙花废弃枝叶的高值化利用,该研究系统探究其黄酮提取物的体外抗氧化活性及体内降血脂效能。通过DPPH自由基、ABTS+自由基及O2-自由基的清除率来研究密蒙花枝叶黄酮的抗氧化性。构建高脂饮食诱导的ICR小鼠模型,设置正常对照组、模型对照组、阳性对照组及提取物低、中、高剂量组,连续干预10周。体外抗氧化实验显示,纯化后的黄酮提取物清除DPPH自由基、ABTS+自由基及O2-自由基的半数抑制浓度分别为31.7、18.4、106.6 μg/mL,其中对ABTS+自由基的清除率优于VC(15.3 μg/mL),对DPPH自由基的清除率优于粗提物(55.1 μg/mL)及VC(35.7 μg/mL)。体内降血脂结果表明,高剂量组可显著抑制小鼠体质量异常增长(P<0.05),其控制体质量效能及对器官指数的改善作用与阳性对照组相当。提取物呈剂量依赖性降低血清总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、低密度脂蛋白胆固醇(low-density lipoprotein cholesterol,LDL-C)水平及动脉硬化指数,缓解高密度脂蛋白胆固醇异常升高;干预8周后,高剂量组血脂指标基本恢复正常,且对LDL-C的下调效果优于阳性对照组。该提取物可提升血清及肝组织总抗氧化能力、超氧化物歧化酶活性,降低丙二醛含量并调节一氧化氮水平,通过维持氧化稳态保护机体;同时降低肝组织 TC、TG 水平,并逆转高脂饮食对3-羟基-3-甲基戊二酸单酰辅酶A还原酶的反馈抑制作用,使该酶活性恢复至正常水平(P<0.05),从而间接调控内源性胆固醇合成。
Abstract:To achieve the high-value utilization of discarded branches and leaves of Buddleja officinalis, the in vitro antioxidant activity and in vivo hypolipidemic efficacy of its flavonoid extract were systematically investigated in this paper. The antioxidant activity of flavonoids from the branches and leaves of Buddleja officinalis was investigated by evaluating their scavenging rates against DPPH radicals, ABTS+ radicals, and O2- radicals. An ICR mouse model induced by a high-fat diet was established, and the mice were divided into a normal control group, a model control group, a positive control group, and low-, medium-, and high-dose extract groups, with continuous intervention for 10 weeks. In vitro experiments showed that the median inhibition concentration values of the purified extract for scavenging DPPH radicals, ABTS+ radicals, and superoxide anion radicals were 31.7, 18.4, and 106.6 μg/mL, respectively. Among them, the scavenging activity against ABTS+ radicals was superior to that of VC (15.3 μg/mL), and the scavenging activity against DPPH radicals was superior to that of the crude extract (55.1 μg/mL) and VC (35.7 μg/mL). In vivo lipid-lowering results showed that, the abnormal increase in body weight of mice was significantly inhibited by the high-dose group(P<0.05), and its efficacy in controlling body weight and improving organ indices was comparable to that of the positive control group. Serum total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), and atherosclerosis index were dose-dependently reduced by the extract, and the abnormal elevation of high-density lipoprotein cholesterol was corrected. After 8 weeks of intervention, the blood lipid indices in the high-dose group basically returned to normal, and the down-regulation effect on LDL-C was superior to that of the positive control group. The total antioxidant capacity and superoxide dismutase activity in serum and liver tissues were enhanced; the malondialdehyde content was decreased, and the nitric oxide level was regulated by the extract, thereby protecting the body by maintaining oxidative homeostasis. Meanwhile,the deposition of TC and TG in liver tissues was reduced, and reversed the feedback inhibition of 3-hydroxy-3-methylglutaryl coenzyme A reductase induced by a high-fat diet, restoring the enzyme activity to near-normal levels (P<0.05), thereby indirectly regulating endogenous cholesterol synthesis.
