中文摘要: 该研究旨在优化玛卡多糖提取工艺参数,并进一步对其抗氧化活性进行初步分析。利用水提醇沉法提取玛卡多糖,采用单因素试验考察提取时间、提取温度、料液比和提取次数对玛卡多糖提取率的影响;以多糖提取率为响应值,运用Box-Behnken设计四因素三水平试验并进行响应面优化试验,确定各影响因素与多糖提取率的关系;以抗坏血酸为阳性对照,通过测定玛卡多糖对DPPH自由基、ABTS+自由基及羟基自由基的清除能力,评估其抗氧化活性。结果表明,玛卡多糖最佳提取条件为提取时间120 min、提取温度70 ℃、料液比1∶30 (g/mL)、提取3次。在此条件下,玛卡多糖提取率达30.38%。抗氧化活性测定结果显示,玛卡多糖具有显著的自由基清除能力,表现出良好的抗氧化潜力。
Abstract:This study aims to optimize the extraction process parameters of maca polysaccharides and preliminarily analyze their antioxidant activity. Maca polysaccharides were extracted by water extraction and ethanol precipitation method. The effects of extraction time, extraction temperature, solid-liquid ratio, and number of extractions on the extraction yield of maca polysaccharides were investigated by single-factor experiments.With the extraction yield of polysaccharides as the response value, a four-factor and three-level experiment was designed using Box-Behnken design, and response surface optimization experiments were conducted to determine the relationship between each influencing factor and the extraction yield of polysaccharides. By using ascorbic acid as the positive control, the antioxidant activity was evaluated by measuring the scavenging ability of maca polysaccharides against DPPH radicals, ABTS+ radicals, and hydroxyl radicals. The results showed that the optimal extraction conditions were as follows: extraction time of 120 min, extraction temperature of 70 ℃, solid-liquid ratio of 1∶30 (g/mL), and three extractions. Under these conditions, the extraction yield of maca polysaccharides reached 30.38%. The antioxidant activity tests showed that maca polysaccharides possessed significant free radical scavenging ability and exhibited good antioxidant potential.
