中文摘要: 为探究山药-黄芪-黄精复合营养羹对免疫力低下模型小鼠肠黏膜屏障损伤的修复作用及机制,采用环磷酰胺(cyclophosphamide,CTX)腹腔注射小鼠构建免疫力低下模型,设置正常组、模型组、复合营养羹高剂量组(21.37 g/kg)、中剂量组(10.67 g/kg)、低剂量组(5.34 g/kg),按组别分别予以等体积生理盐水和复合营养羹溶液灌胃7d,每日1次。通过苏木素-伊红染色观察肠组织病理变化,免疫荧光染色观察小肠闭合小环蛋白-1(zonula occludens-1,ZO-1)、闭锁蛋白(occludin)的阳性定位,实时荧光定量聚合酶链式反应检测肠组织ZO-1、occludin、多聚免疫球蛋白受体(polymeric immunoglobulin receptor,pIgR)的mRNA表达,蛋白质印迹法分别检测ZO-1、occludin、pIgR、分泌型免疫球蛋白A(secretory immunoglobulin A,sIgA)的蛋白表达,采用蛋白酶联免疫吸附测定法检测肠组织的细胞因子白细胞介素 -10、白细胞介素-2、sIgA含量以及肠黏膜黏液中的溶菌酶含量。结果表明,复合营养羹可改善环磷酰胺诱导免疫力低下小鼠的一般情况,促进体质量增长,提高脾脏指数,减轻肠组织病理损伤;可上调小肠上皮紧密连接分子ZO-1和occludin的表达,提高细胞因子白细胞介素-10和白细胞介素-2的含量,增加小鼠溶菌酶、pIgR、sIgA的含量,尤其是以复合营养羹高剂量组尤为明显。综上,复合营养羹能够改善免疫力低下小鼠的肠道黏膜屏障损伤。
Abstract:To explore the repairing effect and underlying mechanism of Chinese yam-Astragalus-Polygonati Rhizoma compound nutritious soup on intestinal mucosal barrier injury in immunosuppressed model mice, an immunocompromised mouse model was established by intraperitoneal injection of cyclophosphamide (CTX). Mice were divided into a normal group, a model group, a high-dose compound nutritious soup group (21.37 g/kg), a medium-dose compound nutritious soup group (10.67 g/kg), and a low-dose compound nutritious soup group(5.34 g/kg). According to group assignment, mice were intragastrically administered equal volumes of normal saline or compound nutritious soup solution once daily for 7 days. Histopathological changes in intestinal tissues were observed by hematoxylin-eosin staining. Immunofluorescence staining was used to determine the positive localization of zonula occludens-1 (ZO-1) and occludin in the small intestine. Real-time quantitative polymerase chain reaction was performed to detect the mRNA expression levels of ZO-1, occludin, and polymeric immunoglobulin receptor (pIgR) in intestinal tissues. Western blot analysis was used to determine the protein expression levels of ZO-1, occludin, pIgR, and secretory immunoglobulin A (sIgA). Enzyme-linked immunosorbent assay (ELISA) was used to measure the levels of cytokines interleukin-10 (IL-10) and interleukin-2 (IL-2), the content of sIgA in intestinal tissues, and the content of lysozyme in intestinal mucosal mucus. The results showed that compound nutritious soup improved the general condition of CTX-induced immunocompromised mice, promoted body weight gain, increased the spleen index, and alleviated pathological damage in intestinal tissues. It also upregulated the expression of the tight junction molecules ZO-1 and occludin in the small intestinal epithelium, increased the levels of cytokines IL-10 and IL-2, and enhanced the activities of lysozyme, pIgR, and sIgA in mice, with the most pronounced effects observed in the high-dose compound nutritious soup group. In conclusion, compound nutritious soup can improve intestinal mucosal barrier injury in immunocompromised mice.
