中文摘要: 该文以薄壳山核桃饼粕和黄豆饼粕(对照)为原料,通过3 种菌种(枯草芽孢杆菌、虫草菌和侧耳菌)固态发酵制备薄壳山核桃蛋白肽,对薄壳山核桃粕蛋白肽的蛋白质浓度、抗氧化活性、乳化性及其制备的微胶囊粉特性进行研究。结果表明:相较于其他菌种,枯草芽孢杆菌发酵薄壳山核桃饼粕制备的蛋白肽样品粗蛋白浓度最高,发酵3 d和9 d时分别为21.27 mg/mL和25.60 mg/mL;经侧耳菌发酵薄壳山核桃饼粕9 d得到的蛋白肽总抗氧化能力和DPPH自由基清除能力最强,总抗氧化能力为12.93 mmol/L FeSO4;DPPH自由基清除能力为0.20 μg Trolox/mL。虫草菌发酵黄豆饼粕得到的蛋白肽乳化性最好,其乳化活性和乳化稳定性在发酵9 d时分别为0.019 m2/g和98.58%。同时采用侧耳菌发酵制备高抗氧化活性蛋白肽包埋薄壳山核桃油制备微胶囊粉,相较于常规壁材(β-环糊精),采用侧耳菌发酵制备的高抗氧化活性蛋白肽作为壁材时,其微胶囊粉的包埋率显著提高至85.28%,且总抗氧化能力(12.93 mmol/L FeSO4)和DPPH自由基清除率(0.20 μg Trolox/mL)均显著优于β-环糊精包埋薄壳山核桃油微胶囊粉(分别为 6.68 mmol/L FeSO4 和 0.17 μg Trolox/mL)。
Abstract:Pecan cake meal and soybean cake meal (as a control) were used as raw materials to prepare pecan protein peptides via solid-state fermentation using three strains (Bacillus subtilis,Cordyceps militaris,and Pleurotus eryngii).The protein concentration,antioxidant activity,and emulsifying properties of the pecan cake meal protein peptides,as well as the characteristics of the resulting microcapsule powder,were investigated.The results showed that,compared with other strains,the protein peptides prepared from pecan cake meal fermented by B.subtilis exhibited the highest crude protein concentration,which reached 21.27 and 25.60 mg/mL at 3 d and 9 d of fermentation,respectively.The protein peptides obtained from pecan cake meal fermented by P.eryngii for 9 d showed the strongest total antioxidant capacity and the DPPH radical scavenging ability.The total antioxidant capacity was 12.93 mmol/L FeSO4,and the DPPH radical scavenging rate was 0.20 μg Trolox/mL.The protein peptides obtained from soybean cake meal fermented by C.militaris exhibited the best emulsifying properties.The emulsifying activity and emulsion stability were 0.019 m2/g and 98.58% at 9 d of fermentation,respectively.In addition,protein peptides with high antioxidant activity prepared by P.eryngii fermentation were used as wall materials to encapsulate pecan oil for the preparation of microcapsule powder.Compared with the conventional wall material (β-cyclodextrin),when protein peptides with high antioxidant activity prepared by P.eryngii fermentation were used as wall materials,the encapsulation efficiency of the microcapsule powder was significantly increased to 85.28%,and both the total antioxidant capacity (12.93 mmol/L FeSO4) and DPPH radical scavenging rate (0.20 μg Trolox/mL) were significantly superior to those of the β-cyclodextrin-encapsulated pecan oil microcapsule powder (6.68 mmol/L FeSO4 and 0.17 μg Trolox/mL,respectively).
