中文摘要: 为提高甜龙竹笋壳的综合开发利用价值,促进甜龙竹产业发展,该研究选取甜龙竹笋壳作为试验材料,利用超声辅助碱法,在单因素试验基础上,通过Box-Behnken响应面设计方法进一步优化甜龙竹笋壳中结合多酚的提取工艺,并对其抗氧化和降血糖活性进行评估。结果显示,最佳提取工艺为碱液浓度8 mol/L、液料比17∶1(mL/g)、超声时间54 min、超声功率200 W,此条件下结合多酚提取量较高,为18.69 mg GAE/g,与预测的结果(18.70 mg GAE/g)高度一致。结合多酚对DPPH·和ABTS+·的清除能力较强,其半数抑制浓度(median inhibition concentration,IC50)分别为0.22、0.020 3 mg/mL,低于游离多酚(对应值分别为0.32、0.023 1 mg/mL)。结合多酚对α-淀粉酶及α-葡萄糖苷酶也具有较强的抑制效果,IC50值分别为0.05、0.16 mg/mL,而游离多酚对应的抑制效果稍弱(IC50值分别为0.06、0.19 mg/mL)。综上所述,甜龙竹笋壳中的结合多酚不仅具备良好的抗氧化活性,还具有优秀的降血糖潜力。
Abstract:Bound polyphenols were obtained from Dendrocalamus hamiltonii shoot shells via ultrasoundassisted alkali extraction to enhance the comprehensive utilization value and advance the D.hamiltonii industry.On a foundation of single-factor experiments,a Box-Behnken response surface design was adopted to optimize the extraction process.Subsequently,the antioxidant capacity and blood glucose-lowering activity of the extracted bound polyphenols were evaluated.The optimal extraction parameters were determined as follows:an alkali concentration of 8 mol/L,a liquid-to-solid ratio of 17∶1 (mL/g),ultrasonic duration of 54 min,and ultrasonic power of 200 W.Under these conditions,the yield of bound polyphenols was the highest (18.69 mg GAE/g),showing excellent agreement with the value (18.70 mg GAE/g) predicted by the regression model.The bound polyphenols demonstrated potent scavenging activities against DPPH· and ABTS+· radicals,with median inhibitory concentration (IC50) values of 0.22 mg/mL and 0.020 3 mg/mL,respectively,outperforming free polyphenols (IC50 values of 0.32 mg/mL and 0.023 1 mg/mL,respectively).Furthermore,the bound polyphenols exhibited strong inhibitory effects on α-amylase and α-glucosidase,with IC50 values of 0.05 mg/mL and 0.16 mg/mL,respectively,whereas free polyphenols showed marginally lower corresponding inhibitory effects,with IC50 values of 0.06 mg/mL and 0.19 mg/mL.In summary,the bound polyphenols derived from D.hamiltonii shoot shells possessed significant antioxidant activity and considerable blood glucose-lowering potential.
