中文摘要: 为探究茶树精油和百里香精油的最佳提取工艺、化学成分和抗氧化、抑菌活性,采用超声波辅助水蒸气蒸馏法提取茶树精油和百里香精油,精油提取率作为优化指标,采用单因素试验和响应曲面法优化两种精油的提取工艺;利用气相色谱-质谱联用法和总抗氧化能力(total antioxidant capacity,T-AOC)检测试剂盒对两种精油进行成分鉴定和体外总抗氧化能力测定,并以3 种常见致病菌为供试菌测定两种精油的抑菌活性。结果表明,茶树精油的最佳提取工艺为盐浓度6.13%、液料比10∶1(mL/g)、提取时间1.1 h,此时精油提取率为(2.53±0.45)%,百里香精油的最佳提取工艺为盐浓度4.18%、液料比15.04∶1(mL/g)、提取时间1.1 h,此时精油提取率为(1.48±0.21)%。茶树精油共鉴定出17 种挥发性化合物,主要挥发性物质是萜烯类和醇类物质,相对百分含量分别为59.27%、37.84%,未检出酚类物质;百里香精油共鉴定出22 种挥发性化合物,主要是烯类、酚类和醇类物质,相对百分含量分别为75.93%、14.03%、6.69%;茶树精油和百里香精油均具有良好的抗氧化活性并呈浓度依赖性,且更高含量的酚类和烯类物质赋予百里香精油更高的抗氧化活性;两种精油的抑菌圈直径介于16.77~26.45 mm,最小抑菌浓度在0.182 3~5.833 3 mg/mL,最小杀菌浓度在0.364 6~11.666 7 mg/mL,表明茶树精油和百里香精油具有显著的广谱抗菌活性。
Abstract:To investigate the optimal extraction processes,chemical compositions,and antioxidant and antibacterial activities of tea tree essential oil and thyme essential oil,ultrasound-assisted water distillation was used to extract the two essential oils.Extraction yield was used as the optimization index,and single-factor experiments combined with response surface methodology were applied to optimize the extraction processes.Gas chromatography-mass spectrometry (GC-MS) and a total antioxidant capacity (T-AOC) assay kit were used to identify the chemical components and determine the in vitro total antioxidant activities of the two essential oils,respectively.The antibacterial activities were evaluated using three common pathogenic bacteria as test strains.The results showed that the optimal extraction conditions for tea tree essential oil were a sodium chloride concentration of 6.13%,a liquid-to-material ratio of 10∶1 (mL/g),and an extraction time of 1.1 h,with an extraction yield of (2.53±0.45)%.The optimal extraction conditions for thyme essential oil were a sodium chloride concentration of 4.18%,a liquid-to-material ratio of 15.04∶1 (mL/g),and an extraction time of 1.1 h,with an extraction yield of (1.48±0.21)%.A total of 17 compounds were identified in tea tree essential oil,and the main volatile components were olefins and alcohols,accounting for 59.27% and 37.84%,respectively,phenolic compounds were not detected.A total of 22 known compounds were identified in thyme essential oil,mainly olefins,phenols,and alcohols,accounting for 75.93%,14.03%,and 6.69%,respectively.Both tea tree essential oil and thyme essential oil exhibited good antioxidant activity with concentration dependence,and the higher contents of phenolic and olefinic compounds conferred stronger antioxidant activity to thyme essential oil.The inhibition zone diameters of the two essential oils ranged from 16.77 mm to 26.45 mm,the minimum inhibitory concentrations ranged from 0.182 3 mg/mL to 5.833 3 mg/mL,and the minimum bactericidal concentrations ranged from 0.364 6 mg/mL to 11.666 7 mg/mL,indicating that tea tree essential oil and thyme essential oil possessed significant broad-spectrum antibacterial activity.
