中文摘要: 采用低温热风干燥、高温热风干燥、真空干燥和真空冷冻干燥4 种方法对米糠不溶性膳食纤维(rice bran insoluble dietary fiber,RBIDF)进行干燥,利用分光光度法测定RBIDF中的游离酚、结合酚含量及抗氧化活性[DPPH·清除能力和亚铁离子还原力(ferric reducing antioxidant power,FRAP)],并采用高效液相色谱对酚类物质进行定性和定量分析,同时采用体外模拟消化的方法比较4 种干燥方式对RBIDF中酚类物质生物可及性的影响。结果表明,4 种干制RBIDF中结合酚含量及其抗氧化活性(DPPH·清除能力和FRAP)均分别高于相应的游离酚。RBIDF中主要游离酚是表儿茶素和香草醛,两者含量之和占游离态单体酚总含量的90%以上;RBIDF中主要结合酚是阿魏酸与对香豆酸,两者含量之和占结合态单体酚总含量的99%以上。从扫描电子显微镜形貌观察和红外光谱分析结果来看,不同干燥方法对RBIDF结构存在一定影响。综上,与热风干燥、真空冷冻干燥相比,真空干燥更有利于RBIDF中酚类物质在干燥过程中的保留和消化过程中的释放,从而使RBIDF保持相对较高的抗氧化活性和生物可及性。
Abstract:Rice bran insoluble dietary fiber (RBIDF) was dried using four methods,namely low-temperature hot air drying,high-temperature hot air drying,vacuum drying,and vacuum freeze drying.The content of free and bound phenolics in RBIDF,as well as their antioxidant activities (DPPH· scavenging capacity and FRAP),was assessed using spectrophotometric techniques.Phenolic compounds were subjected to both qualitative and quantitative analyses using high-performance liquid chromatography (HPLC).Furthermore,the impact of the four drying methods on the bioaccessibility of phenolic compounds in RBIDF was evaluated through in vitro simulated digestion.The results showed that the content of bound phenolics and their antioxidant activities in the four types of dried RBIDF were higher than those of free phenolics.Epicatechin and vanillin were the predominant free phenolics in RBIDF,accounting for over 90% of the total free monomeric phenolics.Ferulic and p-coumaric acids were the predominant bound phenolics,accounting for over 99% of the total bound monomeric phenolics.Scanning electron microscopy and Fourier transform infrared spectroscopy revealed that different drying methods exerted certain impacts on the structure of RBIDF.In conclusion,compared to hot air drying and vacuum freeze drying,vacuum drying was more conducive to the retention of phenolic compounds during drying and their release during digestion,thereby maintaining relatively higher antioxidant activity and bioaccessibility in RBIDF.
