中文摘要: 该研究旨在探索6 种食药两用食用菌水提物的体外生物活性和成效关系。采用苯酚-硫酸法、考马斯亮蓝法、福林酚法和氯化铝法分别测定水提物中的总糖、粗多糖、总蛋白、总酚及总黄酮含量,以α-淀粉酶活性抑制率和葡萄糖透析迟缓指数探究水提物的体外降糖活性,通过2,2'-联氮-双-3-乙基苯并噻唑啉-6-磺酸[2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid),ABTS]自由基、1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基、羟基自由基清除试验和总抗氧化能力试验评估水提物的抗氧化活性,基于RAW264.7细胞评价其潜在的免疫活性。结果表明,鹿茸菌水提物中总糖含量最高,达到398.61 mg/g,黑皮鸡枞菌水提物中总蛋白含量是其他水提物1.8 倍~15.1倍,鸡油菌水提物总酚含量最高(9.84 mg/g)。功能特性方面,香菇水提物的降糖活性最优,抑制α-淀粉酶活性的半抑制浓度值为0.50 mg/mL。鸡油菌的抗氧化活性明显较好,在0.10 mg/mL的浓度下ABTS+自由基清除率高达99.67%。鹿茸菌水提物浓度80 μg/mL时可促进巨噬细胞增殖27.42%,而松茸水提物浓度(1 280 μg/mL)增殖率达29.62%。综上,香菇水提物降糖活性较好,与其粗多糖含量相关;鸡油菌抗氧化活性较强,与其总酚含量相关,后续可进一步开发其相关功能食品。
Abstract:The relationships between in vitro bioactivities and bioactive components of aqueous extracts from six species of edible fungi with medicinal value were explored.The phenol-sulfuric acid method,Coomassie Brilliant Blue method,Folin-Ciocalteu method,and aluminum chloride method were employed to quantify total sugars,crude polysaccharides,total proteins,and total phenols and total flavonoids in the aqueous extracts,respectively.The in vitro glucose-lowering activities of the aqueous extracts were evaluated based on the α-amylase inhibition rate and glucose dialysis retardation index.The antioxidant activities of the aqueous extracts were assessed through 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS),1,1-diphenyl-2-picrylhydrazyl (DPPH),and hydroxyl radical scavenging assays,as well as total antioxidant capacity experiments.RAW264.7 cells were used to evaluate the potential immunomodulatory activities of the aqueous extracts.The results indicated that Lyophyllum decastes aqueous extract exhibited the highest total sugar content (398.61 mg/g).Oudemansiella raphanipies aqueous extract contained the total protein 1.8-15.1 folds that of other aqueous extracts.Cantharellus cibarius aqueous extract showed the highest content of total phenols (9.84 mg/g).Lentinus edodes aqueous extract exerted significant glucose-lowering effect in vitro,with an IC50 value of 0.50 mg/mL in inhibiting α-amylase activity.C.cibarius demonstrated superior antioxidant capacity,achieving an ABTS+ radical scavenging rate of 99.67% at 0.1 mg/mL.L.decastes aqueous extract at low concentrations (80 μg/mL) promoted macrophage proliferation by 27.42%,while Tricholoma matsutake aqueous extract at a high concentration (1 280 μg/mL) increased the macrophage proliferation rate to 29.62%.The good glucose-lowering effect of L.edodes aqueous extract was related to the content of crude polysaccharides,and the strong antioxidant activity of C.cibarius was associated with the content of total phenols.The findings provide a theoretical basis for the development of related functional foods.
