鸡蛋清白蛋白-EGCG非共价物生物利用度及抗氧化活性分析

中文摘要: 混合法制备鸡蛋清白蛋白(egg white albumin,E)-表没食子儿茶素没食子酸酯(epigallocatechin gallate,EGCG)非共价物后,模拟口腔、胃及小肠消化,测定E-EGCG非共价物消化前后的EGCG含量及DPPH自由基、ABTS+自由基和羟自由基清除率,研究EGCG质量浓度对E-EGCG非共价物EGCG结合量、EGCG生物利用度指数、EGCG生物可接受率和抗氧化活性及抗氧化生物利用度指数的影响。结果表明,随着EGCG质量浓度的增加,E-EGCG非共价物中EGCG结合量、EGCG生物利用度指数和体外抗氧化活性均显著增加(P<0.05),当EGCG质量浓度为10.00 mg/mL时达到最高,分别为1.207 mg/mg、0.637、68.31%(DPPH自由基清除率)、89.32%(羟自由基清除率)和84.00%(ABTS+自由基清除率)。与等浓度(1.25 mg/mL)游离EGCG相比,E-EGCG非共价物中的EGCG具有更高的DPPH自由基和羟自由基清除率、生物可接受率,体外消化后产物羟自由基清除率得到显著提高。
Abstract:Egg white albumin (E)-epigallocatechin gallate (E-EGCG) non-covalent complex was prepared by the mixing method;then,the oral,gastric,and small intestinal digestion was simulated to determine the EGCG content and the scavenging rates of DPPH,ABTS+,and hydroxyl radicals before and after digestion of the E-EGCG non-covalent complex.The effect of EGCG mass concentration on the EGCG binding amount,EGCG bioavailability index,EGCG bioaccessibility,antioxidant activity,and bioavailability index of E-EGCG noncovalent complex was studied.The results showed that the EGCG binding amount,EGCG bioavailability index,and in vitro antioxidant activity in the E-EGCG non-covalent complex significantly increased (P<0.05) with the increasing EGCG mass concentration,reaching the maximum values of 1.207 mg/mg,0.637,68.31%(DPPH radical scavenging rate),89.32% (hydroxyl radical scavenging rate),and 84.00% (ABTS+ radical scavenging rate) at an EGCG mass concentration of 10.00 mg/mL.Compared to the free EGCG at the same concentration (1.25 mg/mL),the EGCG in the E-EGCG non-covalent complex exhibited higher DPPH and hydroxyl radical scavenging rates,as well as higher bioaccessibility,and the hydroxyl radical scavenging rate of the digestive products was significantly increased after in vitro digestion.
