中文摘要: 该文采用超声辅助低共熔溶剂(deep eutectic solvent,DES)法对山茱萸中多糖进行提取。通过单因素试验和响应面试验,以粗多糖提取率为指标,优化山茱萸多糖提取工艺,并通过自由基清除试验,对山茱萸多糖抗氧化活性进行研究。结果表明,最佳提取条件为DES含水量53%、超声温度32 ℃、料液比1∶42 (g∕mL)、超声时间30 min,该条件下粗多糖提取率为17.23%,将粗多糖纯化后,对其抗氧化能力进行测定。结果表明,山茱萸多糖对DPPH自由基、ABTS+自由基、羟基自由基均有清除作用,其半抑制浓度(half maximal inhibitory concentration,IC50)分别为0.48、0.51、0.80 mg∕mL。
Abstract:Ultrasound-assisted deep eutectic solvent (DES) extraction was employed to obtain polysaccharides from Corni Fructus.The extraction process was optimized by single-factor experiments and response surface methodology,with the crude polysaccharide extraction rate as the indicator.Furthermore,the antioxidant activity of the polysaccharides was investigated via free radical scavenging assays.The results showed that the optimal extraction conditions were as follows: DES water content of 53%,ultrasonic temperature of 32 ℃,solid-liquid ratio of 1∶42 (g∕mL),and ultrasonic time of 30 min.Under these conditions,the crude polysaccharide extraction rate reached 17.23%.After purification of the crude polysaccharides,their antioxidant activity was determined.The results showed that the polysaccharides from Corni Fructus exhibited scavenging effects on DPPH·,ABTS+· and hydroxyl radicals,with half-maximal inhibitory concentrations (IC50) of 0.48,0.51,and 0.80 mg∕mL,respectively.
