中文摘要: 以藜麦为原料,分别采用碱溶酸沉法和Osborne分级法对其分离蛋白和组分蛋白(白蛋白、球蛋白、谷蛋白)进行提取和分离,探究其热力学特性、二级结构、分子量分布等理化性质,同时测定其乳化特性、起泡能力和持水性及持油性等功能性质。结果显示,4种蛋白质在还原与非还原电泳模式下,其20~50 kDa间的条带展示出明显的不同;在热力学特性方面藜麦谷蛋白具有较高的热稳定性;傅里叶变换红外光谱证实不同蛋白二级结构存在明显不同;分离蛋白具有较多的二硫键和适中的游离巯基;球蛋白具有较高的表面疏水性;各蛋白溶解度在pH值变动中均显示出典型的U型曲线,尤以分离蛋白的溶解特性最为明显。此外,藜麦分离蛋白的持水性和乳化活性指数最好;藜麦球蛋白持油性最佳;藜麦白蛋白起泡性和泡沫稳定性较好。
Abstract:Quinoa was used as raw material for the extraction of protein isolate and separation of component proteins (albumin,globulin,and glutenin) by alkaline solubilization-acid precipitation and Osborne grading methods,respectively.The physicochemical properties such as thermodynamic properties,secondary structure,and molecular weight distribution,as well as the functional properties such as emulsification properties,foaming ability,and water-and oil-holding properties were determined.The results showed that the subunit bands between 20 kDa and 50 kDa of the four proteins were significantly different in reducing and non-reducing electrophoresis modes.Quinoa glutenin had higher thermal stability.Fourier transform infrared spectroscopy confirmed the significant differences in the secondary structures among different proteins.Glutenin had a high amount of disulfide bonds and a moderate amount of free sulfhydryl groups.Globulin had higher surface hydrophobicity.The solubility of the proteins showed typical U-shaped curves with changes in pH.Especially,the solubility characteristics of the protein isolate were the most significant.In addition,quinoa isolate had the best water holding and emulsifying properties.Quinoa globulin had the best oil-holding properties.Quinoa albumin had excellent foaming ability.
