中文摘要: 使用不同浓度乙醇溶液(40%、50%、60%、70%和80%)分级沉淀得到5组山药多糖(P40、P50、P60、P70和P80),对分级多糖的结构特征和降脂活性进行分析与检测。结果表明,山药分级多糖均是由甘露糖、氨基葡萄糖、核糖、鼠李糖、葡萄糖醛酸、半乳糖醛酸、葡萄糖、半乳糖、木糖、阿拉伯糖聚合而成的α-构型低聚糖,具有O—H、C—H、Cpagenumber_ebook=24,pagenumber_book=16O、C—O—C和—NH2等官能团结构。在检测浓度范围内,山药分级多糖呈剂量依赖性体外降脂活性,其中P60的综合降脂能力最强。当浓度为10 mg∕mL时,P60对胰脂肪酶活性抑制率、胆固醇酯酶活性抑制率及胆固醇胶束溶解度抑制率分别为(14.67±0.74)%、(45.21±1.91)%和(48.61±2.63)%,对牛磺胆酸钠和甘氨胆酸钠的结合率分别达到(50.04± 1.23)%和(32.21±1.90)%,具有较好的体外降脂活性。

Abstract:Five groups of yam polysaccharides (P40,P50,P60,P70,and P80) were obtained by graded precipitation using different concentrations of ethanol (40%,50%,60%,70%,and 80%).Then,the structural characteristics and lipid-lowering activity of the graded polysaccharides were analyzed and detected.The results showed that the graded polysaccharides from yams were all α-oligosaccharides polymerized from mannose,glucosamine,ribose,rhamnose,glucuronic acid,galacturonic acid,glucose,galactose,xylose,and arabinose,possessing functional group structures such as O—H,C—H,Cpagenumber_ebook=24,pagenumber_book=16O,C—O—C and—NH2.Within the tested concentration range,the graded polysaccharides from yams exhibited lipid-lowering abilities in a dose-dependent manner in vitro,and P60 possessed the strongest comprehensive lipid-lowering ability.When the concentration was 10 mg∕mL,the inhibition rates of P60 on pancreatic lipase activity,cholesterol esterase activity,and cholesterol micelle solubility reached (14.67±0.74)%,(45.21±1.91)%,and (48.61±2.63)%,respectively,and the binding rates to sodium taurocholate and sodium glycocholate reached (50.04±1.23)%and (32.21±1.90)%,respectively,indicating it has good lipid-lowering activity in vitro.
