中文摘要: 为揭示玫瑰红茶加工过程中优势微生物的分布规律及其对茶品质和安全性的潜在影响,该文对玫瑰红茶5个关键加工阶段(鲜花期、萎凋期、揉捻期、发酵期和干燥期)的细菌和真菌进行培养、分离、菌落形态鉴定,并通过细菌16S rRNA和真菌内转录间隔区(internal transcribed spacer,ITS)序列进行分子生物学鉴定。结果显示,从5个加工阶段中共筛选并鉴定出12株细菌和12株真菌。细菌中芽孢杆菌属(地衣芽孢杆菌和枯草芽孢杆菌)、成团泛菌及表皮葡萄球菌,以及真菌中季也蒙假丝酵母、热带假丝酵母、塔宾曲霉、黑曲霉、球毛壳霉菌均对玫瑰红茶品质形成起到积极作用。玫瑰红茶的独特品质和风味是通过多种微生物的协同作用形成的。同时,该研究针对传统生产工艺存在的生物安全隐患,提出管理建议。
Abstract:Rose black tea was a substitute tea made from rose petals via the processing techniques of black tea. During the production of rose black tea,microorganisms play a crucial role in its quality,functional properties,flavor,and biosafety. The distribution patterns of dominant microorganisms during the processing of rose black tea and their potential impacts on tea quality and safety were studied. Bacteria and fungi were cultured,isolated,and initially identified based on colony morphology during five key processing stages (flower,withering,rolling,fermentation,and drying stages) of rose black tea. Molecular identification was subsequently performed based on bacterial 16S rRNA and fungal internal transcribed spacer (ITS) sequences. A total of 12 bacterial strains and 12 fungal strains were isolated and identified from the five processing stages. The potential roles of these dominant microorganisms in the quality formation of rose black tea were discussed. Bacillus (especially B. licheniformis and B. subtilis),Pantoea agglomerans,and Staphylococcus epidermidis among bacteria,as well as Meyerozyma guilliermondii,Candida tropicalis,Aspergillus tubingensis,Aspergillus niger,and Chaetomium globosum among fungi,contributed positively to the product quality. The unique quality and flavor of rose black tea were formed through the synergistic action of multiple microorganisms. Moreover,management recommendations for rose black tea production considering the potential biosafety hazards in traditional processing methods were proposed.
