胡萝卜非挥发性呈味物质检测方法的优化与应用

中文摘要: 为深入解析胡萝卜风味形成的物质基础,该研究开发一种高效、精准的胡萝卜非挥发性风味物质检测方法。基于气相色谱-质谱联用技术(gas chromatography - mass spectrometry,GC-MS),并结合单因素方差分析,系统比较5种不同的提取方法,最终确定甲醇-氯仿-水(两相)提取法为最优方案,该方法对多数物质均有较好的提取效率,可显著提高氨基酸类物质的含量。此外,通过优化衍生化程序,对比35种衍生化温度组合,最终确定最佳衍生化条件为甲氧胺化50 ℃持续0.5 h,硅烷化70 ℃持续1.5 h。在此条件下,D-果糖、D-葡萄糖、蔗糖、苹果酸等11种关键风味物质的检测含量显著高于其他条件。在此条件下对4个胡萝卜品种的关键风味物质进行检测和分析,揭示可溶性糖是区分不同胡萝卜品种的关键风味成分,并最终通过偏最小二乘判别分析(partial least squares discrimination analysis,PLS-DA)进一步验证该方法的可靠性和稳定性。
Abstract:An efficient and precise method for the determination of non-volatile flavor substances in carrots was developed to deeply analyze the material basis of carrot flavor formation. Gas chromatography-mass spectrometry (GC-MS) was combined with one-way analysis of variance (ANOVA) to systematically compare five different extraction methods. The methanol-water-chloroform (two-phase) extraction method was finally identified as the optimal solution,which showed good extraction efficiency for most substances and significantly improved the content of amino acids. In addition,35 combinations of derivatization temperatures were compared for optimization of the derivatization procedure,and the optimal derivatization conditions were finally determined as follows: methoxylation at 50 ℃ for 0.5 h and silylation at 70 ℃ for 1.5 h. Under the optimal derivatization conditions,the levels of 11 key flavor substances including D-fructose,D-glucose,sucrose,and malic acid were significantly higher than those under other conditions. The key flavor substances of four carrot varieties were determined and analyzed under the optimal derivatization conditions,which revealed that soluble sugars were the key flavor components distinguishing different carrot varieties. Finally,the reliability and stability of the method were verified by partial least squares discriminant analysis (PLS-DA).
