中文摘要: 为研究枸杞多糖(Lycium barbarum polysaccharides,LBP)的抗疲劳作用,该研究以斑马鱼为模型,设置正常对照组、模型对照组、阳性对照组、枸杞水提物与枸杞(Lycium barbarum fruit,LBF)构建的双剂量对照体系实验组[LBP-1(枸杞水提物870 μg/mL)与LBF (2 000 μg/mL)等量多糖组、LBP-2(枸杞水提物2 000 μg/mL)高剂量组]。除正常对照组外,其余各组均通过无水亚硫酸钠建立斑马鱼疲劳模型。各组经相应处理后,测定其乳酸含量、腺嘌呤核苷三磷酸(adenosine triphosphate,ATP)含量、活性氧(reactive oxygen species,ROS)浓度及运动距离等生物指标。结果表明,与模型对照组相比,LBP-1、LBP-2及LBF处理组的乳酸含量和ROS浓度均显著降低、ATP含量及运动距离显著提升(P<0.05),其中LBP效果显著,起主导作用。在其作为疲劳改善剂使用时,适宜剂量为0~870 μg/mL。该研究对LBP的抗疲劳作用进行多重分析,为其在功能食品开发中的应用提供了参考。
Abstract:To investigate the anti-fatigue effects of Lycium barbarum polysaccharides (LBP),zebrafish were used as a model organism. The experimental design included a normal control group,a model control group,a positive control group,and a double-dose control system experimental group constructed using L. barbarum aqueous extract and L. barbarum fruit (LBF). The latter comprised an equivalent polysaccharide group treated with LBP-1 (aqueous extract of L. barbarum,870 μg/mL) and LBF (2 000 μg/mL),and a high-dose group treated with LBP-2 (aqueous extract of L. barbarum,2 000 μg/mL). Except for the normal control group,the fatigue model in zebrafish was established using anhydrous sodium sulfite. After the corresponding treatments,biological indicators including lactic acid content,adenosine triphosphate (ATP) content,reactive oxygen species (ROS) concentration,and locomotor distance were measured in each group. The results showed that,compared with the model control group,lactic acid content and ROS concentration were significantly reduced,while ATP content and locomotor distance were significantly increased in the LBP-1,LBP-2,and LBF-treated groups (P<0.05). Among these,LBP exhibited a more pronounced effect and played a dominant role.When used as a fatigue-ameliorating agent,the appropriate dose of LBP was 0-870 μg/mL. This study provides a multi-dimensional evaluation of the anti-fatigue effects of LBP and offers reference data for its application in the development of functional foods.
