中文摘要: 探讨黄芪多糖(Astragalus polysaccharides,APS)对阿尔茨海默病(Alzheimer′s disease,AD)模型小鼠的影响及作用机制。采用酶法提取APS后测定其总糖、蛋白质、糖醛酸含量,并进行傅里叶变换红外光谱(Fourier transforminfrared spectroscopy,FT-IR)分析。采用氯化铝(AlCl3)联合D-半乳糖(D-galactose,D-gal)诱导AD小鼠模型,灌胃给予APS,Morris水迷宫实验进行行为学检测;酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)测定小鼠脑组织中超氧化物歧化酶(superoxide dismutase,SOD)活性和丙二醛(malondialdehyde,MDA)含量;对脑组织进行苏木精-伊红染色(hematoxylin and eosin stain,HE)及尼氏染色病理分析;Western blot法测定Kelch样环氧氯丙烷相关蛋白-1(Kelch-like ECH-associated protein 1,Keap1)、核因子E2相关因子2(nuclear factor erythroid 2-related factor 2,Nrf2)和β-淀粉样蛋白(amyloid-beta,Aβ)表达。结果表明,APS的总糖含量为(55.76±1.88)%,蛋白质含量为(4.87±0.42)%,糖醛酸含量为(5.85±0.34)%;红外光谱结果显示APS存在多糖特征吸收峰。药理结果表明,APS能够有效缓解AD小鼠记忆障碍,增强记忆力,抑制MDA分泌,提高SOD活性,减轻AD小鼠脑组织的损伤,降低脑组织Keap1、Aβ的蛋白表达,提高Nrf2的蛋白表达。APS具有潜在的缓解AD的作用,其机制可能与激活Keap1/Nrf2/Aβ通路的表达相关。
Abstract:The effects and mechanisms of Astragalus polysaccharides (APS) on Alzheimer′s disease (AD)mouse model were investigated. Following enzymatic extraction,the total glucose,protein,and glucuronic acid content of APS was determined,and Fourier transform-infrared (FT-IR) spectroscopy analysis was conducted. The mouse model of AD was induced by AlCl3 combined with D-galactose. APS was administered by gavage,and Morris water maze test was carried out to examine mouse behavior. The superoxide dismutase(SOD) and malondialdehyde (MDA) levels in mouse brain tissue were measured by enzyme-linked immunosorbent assay (ELISA). Pathological changes in the brain tissue were revealed by hematoxylin and eosin stain(HE) staining and Nissl staining. The expression of Kelch-like ECH-associated protein 1 (Keap1),nuclear factor erythroid 2-related factor 2 (Nrf2),and amyloid-beta (Aβ) was determined by Western blot. The results indicated that APS exhibited the sugar content of (55.76±1.88)%,the protein content of (4.87±0.42)%,and the glucuronic acid content of (5.85±0.34)%. The FT-IR spectroscopy results revealed characteristic absorption peaks indicative of polysaccharides in APS. APS effectively alleviated memory impairment and improved memory function in AD mice. Moreover,it reduced brain tissue damage by inhibiting MDA secretion and enhancing SOD activity. From a mechanism perspective,APS downregulated the expression of Keap1 and Aβ,while upregulating the expression of Nrf2 in the brain tissue. In conclusion,APS possessed potential effects in alleviating AD by activating the Keap1/Nrf2/Aβ pathway.
