3种药用植物发酵提取物的抗氧化活性及应用

中文摘要: 以白茅根、芦根、大麦苗3种药食同源材料为研究对象,通过微生物发酵获得其发酵提取物,并分析发酵提取物的抗氧化活性,通过应用到曲奇饼干分析其抗糖化作用。通过使用公认安全(generally recognized as safe,GRAS)细菌解淀粉芽孢杆菌IT-45发酵白茅根、芦根、大麦苗,冷冻干燥后收集发酵提取物,测定发酵提取液中的成分组成及其含量,利用紫外吸收光谱和傅里叶变换红外光谱法对发酵提取物进行结构表征,研究其体外抗氧化活性。结果表明,白茅根、芦根、大麦苗发酵提取物中总糖含量分别为45.10%、21.81%和16.42%;蛋白质含量分别为6.40%、11.28%和6.36%,酚类物质含量分别为14.90%、29.40%和29.89%。体外抗氧化活性试验表明,芦根发酵提取物与大麦苗发酵提取物对ABTS+、DPPH、羟基自由基清除能力与总还原能力均优于白茅根发酵提取物。添加大麦苗发酵提取物的曲奇饼干与添加白茅根发酵提取物的曲奇饼干对荧光晚期糖基化终末产物的抑制率结果相似,而添加芦根发酵提取物的曲奇饼干抑制率最高,为16.66%,表明3种药食同源物质的发酵提取物具有良好的抗氧化活性和抗糖化作用。

Abstract:Three medicinal and edible homologous materials were taken as research subjects,namely roots of Imperata cylindrica var. major,roots of Phragmites communis Trin.,and Hordeum vulgare L. The fermented extracts of three materials were obtained by microbial fermentation. These fermented extracts were subject to antioxidant activity and antiglycation analysis by adding them to cookies. The generally recognized as safe(GRAS) bacterium Bacillus amyloliquefaciens IT-45 was used to ferment the three materials,with fermented extracts collected by freeze drying. The composition and content of the components in the fermented extract were determined. The structure of the fermented extracts was characterized by UV and Fourier transform infrared spectroscopy. In vitro antioxidant activity was also investigated. The results showed that the total sugar content in the fermented extracts of the three materials was 45.10%,21.81%,and 16.42%,respectively. The protein content was 6.40%,11.28%,and 6.36%,respectively. The phenolic content was 14.90%,29.40%,and 29.89%,respectively. In vitro antioxidant activity study showed that the scavenging ability and total reducing power of roots of P. communis,and H. vulgare against ABTS+,DPPH,and hydroxyl radicals were superior to those of roots of I. cylindrica. For inhibitory rate on fluorescent advanced glycation end products (AGEs),similar results were found in cookies added with fermentation extracts of roots of I. cylindrica and H. vulgare,while the highest inhibitory rate of 16.66% was observed in cookies added with fermentation extracts of roots of P.communis. These findings indicated favorable antioxidant activity and antiglycation efficacy in the fermentation extracts of the three medicinal and edible homologous materials.
