铁皮石斛-地黄对2型糖尿病小鼠的降糖作用

中文摘要: 该文比较鲜、干铁皮石斛-地黄对2型糖尿病小鼠的降糖作用。采用高脂高糖饲料结合腹腔注射链脲佐菌素的方法,建立2型糖尿病小鼠模型,将糖尿病小鼠随机分为模型组、二甲双胍阳性对照组、鲜铁皮石斛-地黄高、低剂量组、干铁皮石斛-地黄高、低剂量组。灌胃期间,监测小鼠体质量及空腹血糖值的变化,灌胃结束后,测定小鼠血清生化指标并观察肝脏、胰腺病理组织切片。结果显示:给药组中仅鲜铁皮石斛-地黄高剂量组小鼠体质量显著增长(P<0.05),增长率为1.59%;与模型组相比,铁皮石斛-地黄各给药组小鼠空腹血糖值从灌胃第4周开始均显著降低(P<0.05,P<0.01),以鲜铁皮石斛-地黄高剂量组降血糖效果最显著。铁皮石斛-地黄各给药组总胆固醇、甘油三酯水平均显著降低(P<0.05,P<0.01);鲜铁皮石斛-地黄组低密度脂蛋白胆固醇水平显著降低(P<0.05,P<0.01),而高密度脂蛋白胆固醇水平显著升高(P<0.05,P<0.01);鲜铁皮石斛-地黄组胰岛素水平显著降低(P<0.01);铁皮石斛-地黄各给药组胰岛素抵抗指数均显著降低(P<0.05)。组织病理学结果显示,各给药组均可不同程度修复小鼠肝脏、胰腺细胞功能损伤。综上,铁皮石斛-地黄可有效降低2型糖尿病小鼠血糖,改善糖脂代谢和胰岛素抵抗,其中鲜铁皮石斛-地黄作用效果更佳。
Abstract:This study compared the hypoglycaemic effects of fresh and dried D.officinale-R.glutinosa on type 2 diabetic mice.A high-fat and high-sugar diet combined with an intraperitoneal injection of streptozotocin was used to establish a mouse model of type 2 diabetes mellitus.The diabetic mice were randomly divided into a model group,a metformin-positive control group,high-and low-dose groups of fresh D.officinale-R.glutinosa,and high-and low-dose groups of dried D.officinale-R.glutinosa was set up simultaneously.During the gavage procedure,changes in body mass and fasting blood glucose were monitored.Following the gavage,serum biochemical indexes were measured,and pathological histological sections of the liver and pancreas were observed.The results showed that only the high-dose group of fresh D.officinale-R.glutinosa had a significant increase in body weight(P<0.05),with a growth rate of 1.59%.Compared with the model group,the fasting blood glucose levels of mice in each D.officinale-R.glutinosa group were significantly lower from the 4th week of gavage(P<0.05,P<0.01),and the high-dose group of fresh D.officinale-R.glutinosa had the most significant hypoglycaemic effect.The total cholesterol and triglyceride levels were significantly reduced in all the D.officinale-R.glutinosa group(P<0.05, P<0.01).In the fresh D.officinale-R.glutinosa group,low-density lipoprotein cholesterol levels were significantly reduced(P<0.05, P<0.01),while high-density lipoprotein cholesterol levels were significantly increased(P<0.05, P<0.01).Insulin levels in the fresh D.officinale-R.glutinosa group were significantly reduced(P<0.01).The insulin resistance index in each treatment group of D.officinale-R.glutinosa was significantly reduced(P<0.05).Histopathological results showed that all treatment groups could repair the functional damage of liver and pancreatic cells in mice to varying degrees.In conclusion,D.officinale-R.glutinosa has been demonstrated to effectively reduce blood glucose and improve glucose-lipid metabolism and insulin resistance in type 2 diabetic mice,and the fresh D.officinale-R.glutinosa showed the most beneficial effect.
