中文摘要: 为了解不同种类预制菜的微生物菌落特征,对含鱼肉、虾肉、鸡肉、猪肉的4类预制菜,利用基质辅助激光解吸电离飞行时间质谱(matrix-assisted laser desorption/ionization time-of-flight mass spectrometry,MALDI-TOF MS)法分析不同种类预制菜微生物检出情况和菌落特征。结果表明,共检测出可信水平的微生物258株共86种,微生物鉴定率为94.51%。其中,检出菌株数量前3位的微生物分别为嗜根库克菌28株、解酪巨型球菌28株和加氏乳球菌10株,占比分别为10.85%、10.85%、3.88%。部分微生物如解酪巨型球菌在4类预制菜中均有检出。不同种类的预制菜的微生物菌群分布和数量均有所差异。鱼肉类预制菜的微生物检测出数量和种类要高于另3类预制菜。屎肠球菌仅在虾肉类预制菜中检出,产吲哚金黄杆菌仅在鸡肉类预制菜中检出。该研究基于MALDI-TOF MS技术分析不同种类预制菜的微生物菌落特征,可作为国家标准方法的补充,为预制菜食品安全风险监测、预警及微生物相关标准的制定提供参考依据。

Abstract:To understand the microbial colony characteristics of different types of precooked dishes,four categories of commercially available precooked dishes with fish,shrimp,chicken,and pork were analyzed using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF MS)to determine microbial detection status and colony features.Results showed that a total of 258 credible microbial strains belonging to 86 species were detected,with an identification rate of 94.51%. Among them,the top three microorganisms in terms of strain count were Kocuria rhizophila(28 strains),Macrococcus caseolyticus(28 strains),and Lactococcus garvieae(10 strains),accounting for 10.85%,10.85%and 3.88%,Some microorganisms,such as Macrococcus caseolyticus,were detected in all four types of precooked dishes. The distribution and quantity of microbial flora varied among different precooked dishes. Fish-based precooked dishes had a higher number and variety of detected microorganisms than the other three categories. Enterococcus faecium was only detected in shrimp-based precooked dishes,while Chryseobacterium indologenes was exclusively found in chicken-based ones.This study analyzed the microbial colony characteristics of different precooked dishes using MALDI-TOF MS,which could serve as a supplement to national standard methods and provide a reference for food safety risk monitoring,early warning,and the formulation of microbe-related standards for precooked dishes.
