中文摘要: 以暗褐网柄牛肝菌子实体为原料,采用热水浸提法提取暗褐网柄牛肝菌多糖(Phlebopus portentosus polysaccharide,PBP),对多糖(PBP)及多糖组分(PBP-I)的结构进行表征,并对PBP及PBP-I的体外抗氧化活性进行研究。结果表明,PBP中总糖含量为70.97%,蛋白质含量为8.14%,葡萄糖醛酸含量为5.60%,还原糖含量为4.16%。紫外光谱扫描结果显示PBP及PBP-I在200 nm波长附近有强吸收峰,符合多糖的紫外吸收特征。PBP及PBP-I由甘露糖、半乳糖醛酸、葡萄糖、半乳糖和岩藻糖以不同物质的量比组成。PBP的分子量分布在4.54×107、1.41×107、6.63×105 Da;PBP-I的分子量分布集中在1.20×107 Da,所占比例为90.4%。红外光谱结果表明,PBP及PBP-I是存在β-糖苷键的吡喃糖。刚果红试验结果表明,PBP含有三螺旋结构。X-射线衍射分析表明,PBP是一种无定型结构。扫描电子显微镜结果显示PBP表面呈黏附团聚的不规则片状,PBP-I呈现不规则纤维丝状聚集。PBP和PBP-I的体外抗氧化活性测定结果表明,PBP和PBP-I清除DPPH自由基、ABTS+自由基的半数抑制浓度(median inhibition concentration,IC50)值分别为3.08、2.82 mg/mL和1.21、2.65 mg/mL,其还原铁离子的半数效应浓度值分别为5.30、18.15 mg/mL。
Abstract:Phlebopus portentosus polysaccharide(PBP)was extracted from the fruiting bodies by the hot water extraction method. The structures of PBP and PBP-I were characterized,and the in vitro antioxidant activities of PBP and PBP-I were studied.The results showed that PBP had the total sugar content of 70.97%,the protein content of 8.14%,the glucuronic acid content of 5.60%,and the reducing sugar content of 4.16%. UV spectral scanning showed strong absorption peaks of PBP and PBP-I around a wavelength of 200 nm,which was consistent with the UV absorption characteristics of polysaccharides.PBP and PBP-I were composed mannose,galacturonic acid,glucose,galactose,and fucose at different ratios.The molecular weight distribution of PBP encompassed 4.54×107,1.41×107 Da,and 6.63×105 Da. The molecular weight of PBP-I was mainly 1.20×107 Da,accounting for 90.4%.The infrared spectroscopy results indicated that PBP and PBP-I contained pyranose with βglycosidic bonds.The results of the Congo Red test indicated that PBP contained a triple helix structure.X-ray diffraction analysis showed that PBP presented an amorphous structure. Scanning electron microscopy showed that the surface of PBP exhibited irregular sheet-like adhesion and aggregation,while PBP-I exhibited irregular fibrous filamentous aggregation. The in vitro antioxidant activity assays of PBP and PBP-I showed that the median inhibition concentration(IC50)values of PBP and PBP-I were 3.08 mg/mL and 2.82 mg/mL for scavenging DPPH radicals and 1.21 mg/mL and 2.65 mg/mL for scavenging ABTS+ radicals,respectively. In addition,PBP and PBP-I showed the median effect concentrations of 5.30 mg/mL and 18.15 mg/mL,respectively,for reducing iron ions.
