中文摘要: 该文通过研究后生元Postbio-YDFF对内氏放线菌的抑菌能力、生物膜损伤以及基因变化,探究后生元Postbio-YDFF对内氏放线菌的抑菌特性及抑菌机制。结果表明:后生元Postbio-YDFF原液对内氏放线菌有显著的抑菌活性,抑菌圈直径为(16.7±0.5)mm,并能显著抑制内氏放线菌生物膜的形成,抑制率达到80.02%,同时破坏菌体的完整性,导致胞内DNA泄漏;通过转录组学分析发现,经后生元Postbio-YDFF处理后,腺嘌呤核苷三磷酸(adenosine triphosphate,ATP)合成(CydA和CydB)、糖运输与碳代谢(PtsH、PtsI和crr)、生物膜(LuxS)和RNA降解(Rho、RNaseJ、DnaK和GroEL)等相关基因显著下调,细胞壁(ALR)和DNA复制(DnaX)等相关基因显著上调,表明后生元Postbio-YDFF通过减少ATP合成、降低碳代谢转换速率、破坏生物膜及干扰遗传信息的表达等途径抑制内氏放线菌生长。
Abstract:This study revealed the antibacterial characteristics and mechanism of postbiotic Postbio-YDFF against Actinomyces naeslundii by investigating the antibacterial ability,biofilm damage,and genetic changes.The results showed that postbiotic Postbio-YDFF had significant antibacterial activity against Actinomyces naeslundii,with a diameter of(16.7±0.5)mm,and it could significantly inhibit the formation of Actinomyces naeslundii biofilm,with an inhibition rate of 80.02%.At the same time,it disrupted the integrity of the bacterial body,leading to intracellular DNA leakage. Through transcriptome analysis,it was found that after treatment with postbiotic Postbio-YDFF,genes related to ATP synthesis(CydA and CydB),sugar transport and carbon metabolism(PtsH,PtsI,and crr),biofilm(LuxS),and RNA degradation(Rho,RNaseJ,DnaK,and GroEL)were significantly downregulated,while genes related to the cell wall(ALR)and DNA replication(DnaX)were significantly upregulated. It indicated that postbiotic Postbio-YDFF inhibited the growth of Actinobacter naeslundii by reducing ATP synthesis and carbon metabolism conversion rate,destroying biofilms,and interfering with the expression of genetic information.
