牡丹籽油提取工艺优化及其体外抗氧化活性分析

中文摘要: 以油用牡丹籽为原料,牡丹籽出油率为考察指标,通过单因素试验和正交试验优化超声辅助水酶法提取牡丹籽油的工艺并探讨牡丹籽油的品质及体外抗氧化活性。结果表明,最优提取工艺为料液比1∶6(g/mL)、木瓜蛋白酶添加量5 000 U/g、酶解pH5.0、酶解温度50.0 ℃、酶解时间4 h。在此条件下,牡丹籽出油率为21.04%。理化检测结果表明,采用超声辅助水酶法制备的牡丹籽油,感官表现为澄清透明、无可见杂质、无异常异味;折光指数(n20)为1.42±0.11;相对密度(d20)为0.92±0.02;酸值为(1.43±0.35)mg/g;碘值为(17.63±1.12)g/kg;皂化值为(193.22±1.43)mg/g;过氧化值为(0.87±0.16)g/kg。各项理化指标均符合GB/T 5492—2008《粮油检验粮食、油料的色泽、气味、口味鉴定》中的要求,品质优异。体外抗氧化试验结果显示,牡丹籽油对1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基、羟基自由基和2,2'-联氮双(3-乙基苯并噻唑啉-6-磺酸)[2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid)cation,ABTS]阳离子自由基的清除率最高分别可达65.29%、55.47%和96.71%,证实其具备良好的体外抗氧化活性。
Abstract:Taking oil-bearing peony seeds as raw material and oil yield as the evaluation index,this study optimized the ultrasonic-assisted aqueous enzymatic extraction(UAAEE)process of peony seed oil through singlefactor experiments and orthogonal experiments.It also systematically investigated the physicochemical quality and in vitro antioxidant activity of the extracted oil.The optimal extraction conditions were determined as follows:solid-liquid ratio of 1∶6(g/mL),papain dosage of 5 000 U/g,enzymatic hydrolysis pH of 5.0,enzymatic hydrolysis temperature of 50.0 ℃ and enzymatic hydrolysis time of 4 h.Under these optimized parameters,the peony seed oil yield reached 21.04%.Physicochemical analysis results indicated that the peony seed oil prepared by UAAEE exhibited sensory characteristics of being clear and transparent,free of visible impurities,and abnormal odors.Its key physicochemical indices were determined as follows:refractive index(n20)of 1.42±0.11,relative density(d20)of 0.92±0.02,acid value of(1.43±0.35)mg/g,iodine value of(17.63±1.12)g/kg,saponification value of(193.22±1.43)mg/g,and peroxide value of(0.87±0.16)g/kg.All the aforementioned indices fully complied with the requirements of relevant national standards,including GB/T 5492—2008 Inspection of grain and oils-Identification of colour,odour and taste of grain and oilseeds,demonstrating the excellent quality of the oil.In vitro antioxidant activity assays showed that the peony seed oil achieved maximum scavenging rates of 65.29%,55.47%,and 96.71% against 1,1-diphenyl-2-picrylhydrazyl(DPPH)radicals,hydroxyl radicals,and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid)cation(ABTS)cation radicals,respectively.These results confirm that the peony seed oil extracted by UAAEE possesses favorable in vitro antioxidant activity.
