中文摘要: 该研究以雪梨、南果梨为主要原料,辅以陈皮、山楂及玫瑰茄,研制一款促消化的复合梨汤饮料。通过单因素试验与正交试验对配方及工艺进行优化,以感官评分为主要评价指标,结合总酚含量、总黄酮含量及抗氧化能力测定结果,确定最佳工艺参数。运用网络药理学探究该饮料促消化的药理作用,筛选并预测促消化活性成分的潜在靶点,重点分析与功能性消化不良相关的靶点。结果表明,复合梨汤饮料的最佳工艺条件为炖煮时间30 min,陈皮、山楂和玫瑰茄添加量分别为2.0%、1.4%和0.6%,此时复合梨汤饮料的总酚含量94.75 mg/L、总黄酮含量112.5 mg/L、1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基清除率89.5%、羟基自由基清除率76.4%、铁离子还原能力0.82 。复合梨汤饮料共筛选出592个有效成分和310个共同靶点。磷脂酰肌醇3-激酶调节亚基1(phosphoinositide 3-kinase regulatory subunit 1,PIK3R1)、蛋白激酶B(protein kinase B,AKT1)、表皮生长因子受体(epidermal growth factor receptor,EGFR)、非受体酪氨酸激酶(steroid receptor coactivator,SRC)、丝裂原活化蛋白激酶激酶1(mitogen-activated protein kinase kinase 1,RAF1)等是复合梨汤饮料治疗功能性消化不良的核心靶点。主要参与的信号通路有磷脂酰肌醇-3-激酶-蛋白激酶B(PI3K-AKT1)通路、脂质与动脉粥样硬化通路和晚期糖基化终末产物及其受体(AGE-RAGE)信号通路。陈皮山楂玫瑰茄复合梨汤饮料治疗功能性消化不良的作用机制可能与改善胃肠动力功能有关。
Abstract:This study utilized snow pears and Nanguo pears as the main raw materials,supplemented with tangerine peel,hawthorn and roselle,to develop a compound pear soup beverage with digestion promoting effect.The formulation and processing parameters were optimized through single-factor experiments and orthogonal design experiments.By employing sensory evaluation scores as the principal assessment criteria,and combining with quantitative analysis of total phenol content,total flavonoid content,and antioxidant capacity,the optimal processing conditions were determined.To investigate the pharmacological effects of this beverage in promoting digestion,network pharmacology approaches were implemented to identify and predict the potential targets of digestion promoting active ingredients,with particular emphasis on the targets associated with functional dyspepsia (FD).The results showed that the optimal processing conditions for the compound pear soup beverage were as follows:a stewing duration of 30 minutes,with tangerine peel,hawthorn and roselle additions at 2.0%,1.4% and 0.6%,respectively.Under these conditions,the total phenol content of the compound pear soup beverage was 94.75 mg/L,the total flavonoid content was 112.5 mg/L,the 1,1-diphenyl-2-picrylhydrazyl(DPPH) free radical scavenging rate was 89.5%,the hydroxyl radical scavenging rate was 76.4%,and the ferric ion reducing ability was 0.82.A total of 592 active ingredients and 310 common targets were identified in compound pear soup beverage.Phosphatidylinositol 3-kinase regulatory subunit 1(PIK3R1),protein kinase B(AKT1),epidermal growth factor receptor (EGFR),steroid receptor coactivator (SRC),mitogen-activated protein kinase kinase 1 (RAF1) and other targets were the core targets of the compound pear soup beverage for treating FD.The predominant signaling pathways implicated in these mechanisms comprised the phosphatidylinositol 3-kinase-protein kinase B(PI3K-AKT1) pathway,lipid metabolism and atherosclerosis pathway,and advanced glycation end products and receptor for advanced glycation end prodncts(AGE-RAGE) signaling pathway.The mechanism of the compound pear soup beverage with tangerine peel,hawthorn and roselle in treating FD might be principally mediated through the enhancement of gastrointestinal function.
