中文摘要: 采用DPPH和ABTS法,研究红石榴浓缩粉(HSL)、苹果提取物(PG)、红葡萄酵母复合粉(HPT)单一组分,以及通过混料设计不同配比组合物的体外抗氧化活性,并通过联用指数(combination index,CI)法评价其协同作用关系。结果表明,单一组分HSL的DPPH自由基和ABTS+自由基清除能力显著高于其他2个单一组分(P<0.05),清除率半数抑制浓度(half maximal inhibitory concentration,IC50)值分别为(7.73±0.66)、(5.74±0.06)μg/mL,但与对照组VC间无明显差异,PG的DPPH自由基清除能力[IC50值为(55.59±3.66)μg/mL]高于HPT[IC50值为(84.15±21.31)μg/mL](P>0.05),但其ABTS+自由基清除能力[IC50值为(32.50±0.28)μg/mL]显著低于HPT[IC50值为(9.16±0.20)μg/mL](P<0.05)。相较于单一组分,复配组分DPPH自由基和ABTS+自由基清除能力IC50值均显著降低,抗氧化活性均显著增加,其中HPT、PG、HSL质量比为15∶15∶70的复配组分(HPH_15∶15∶70)IC50值最低,具有最强的自由基清除活性,且相互作用指数为0.74,表现为强协同作用关系。不同比例复配组分的IC50值与其相互作用指数呈正相关。除HPH_70∶20∶10与HPH_10∶70∶20外,剩余3组的DPPH自由基和ABTS+自由基清除能力的IC50值呈正相关,相关系数为0.863。综上,HSL、PG以及HPT的复配组分在清除DPPH自由基和ABTS+自由基方面能够发挥较好的协同作用,HPH_15∶15∶70活性最强。
Abstract:The in vitro antioxidant activities of pomegranate concentrated powder (HSL),apple extract (PG),red grape yeast compound powder (HPT),and their combinations in different ratios through mixture design were investigated.The 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay and the 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonicacid) diammonium salt(ABTS) free radical scavenging assay were used as the main indicators of antioxidant activity.The synergistic effects of the composite antioxidants were evaluated via the combination index approach.The results showed that HSL exhibited significantly higher DPPH and ABTS+ free radical scavenging rates than PG and HPT (P<0.05),with the half-maximal inhibitory concentrations (IC50) of(7.73±0.66) μg/mL and(5.74±0.06) μg/mL,respectively,which had no significant differences from those of the positive control (VC).PG had higher DPPH free radical scavenging activity [IC50=(55.59±3.66) μg/mL vs.IC50=(84.15±21.31) μg/mL,P>0.05] and lower ABTS+ free radical scavenging activity [IC50=(32.50±0.28) μg/mL vs.IC50=(9.16±0.20) μg/mL,P<0.05] than HPT.Compared with single components,the combinations showed decreases in IC50 values of DPPH and ABTS+ free radical scavenging,demonstrating significantly improved antioxidant activities.The combinations of HPT,PG,and HSL at a mass ratio of 15∶15∶70 (HPH_15∶15∶70) had the lowest IC50 value and the interaction index of 0.74,which indicated the highest radical scavenging activities and the strongest synergistic antioxidant effect,respectively.In addition,there was a positive correlation between the IC50 and the interaction index of combinations at different ratios.Except HPH_70∶20∶10 and HPH_10∶70∶20,the remaining three groups showed a strong correlation (correlation coefficient of 0.863) between the IC50 values for DPPH and ABTS+ free radical scavenging activities.Therefore,the combinations of HSL,PG and HPT exhibited synergistic effects in the DPPH and the ABTS+ free radical scavenging activities,and HPH_15∶15∶70 had the strongest activities.
