酶法制备巴旦木肽的工艺优化及其体外抗氧化、乙酰胆碱酯酶抑制活性

中文摘要: 以巴旦木为原料,通过胰酶与纤维素酶(4∶1,质量比)对巴旦木蛋白进行酶解,以水解度和肽得率为指标,在单因素试验的基础上,通过响应面试验确定巴旦木蛋白酶解的最佳工艺条件为底物浓度4.8%、胰酶添加量14%、酶解时间8 h、酶解温度54 ℃,此条件下巴旦木蛋白的水解度为(55.22±0.56)%,肽得率为(77.92±0.92)%。利用超滤、葡聚糖凝胶 G-25柱层析对巴旦木蛋白酶解肽进行分离纯化,以乙酰胆碱酯酶抑制率为指标,筛选得到具有乙酰胆碱酯酶抑制活性的巴旦木肽(<3 kDa),其DPPH自由基、羟自由基和超氧阴离子自由基清除能力的半数抑制浓度(median inhibition concentration,IC50)分别为5.11、4.34、28.74 mg/mL,总抗氧化能力为1.23 mmol/L。结果表明,具有乙酰胆碱酯酶抑制活性及抗氧化活性的巴旦木肽组分(<3 kDa)分子量主要集中于1.0 kDa以下,占比62.73%。
Abstract:Almond protein was subjected to enzymatic hydrolysis with trypsin and cellulase (4∶1,mass ratio).With the degree of hydrolysis (DH) and peptide yield as key indicators,response surface methodology was employed to optimize the enzymatic hydrolysis conditions based on single-factor experiments.The enzymatic hydrolysis conditions were optimized as follows:substrate concentration of 4.8%,trypsin addition of 14%,hydrolysis duration of 8 h,and hydrolysis temperature of 54 ℃.Under these conditions,the DH was (55.22±0.56)%and the peptide yield reached (77.92±0.92)%.The hydrolysates were separated and purified through ultrafiltration and Sephadex G-25 column chromatography.Almond peptides with acetylcholinesterase (AChE) inhibitory activity and molecular weights less than 3 kDa were screened out with the AChE inhibition rate as the screening criterion.The median inhibition concentration values of the peptides for scavenging DPPH radicals,hydroxyl radicals,and superoxide anion radicals were 5.11,4.34,and 28.74 mg/mL,respectively,and the total antioxidant capacity was determined to be 1.23 mmol/L.The results indicated that the almond peptide fraction(<3 kDa),which demonstrated both AChE inhibitory and antioxidant activities,primarily consisted of peptides with molecular weights below 1 kDa,which accounted for 62.73% of the fraction.
