中文摘要: 以牛眼肉为原料,使用超高效液相色谱-串联质谱(ultra-high performance liquid chromatography tandem mass spectrometry,UHPLC-MS/MS)对4 ℃下贮藏的牛眼肉进行非靶向代谢组学分析。结果表明,从牛眼肉中共鉴定出914种代谢物,主要包括脂质及类脂分子、有机酸及其衍生物和有机杂环化合物等。通过随机森林方法筛选出40种潜在的差异代谢产物,经过聚类分析以及通路富集分析得出这些差异代谢物主要参与氮代谢、谷胱甘肽代谢、甘油酸和二羧酸代谢、精氨酸生物合成、嘧啶代谢以及戊糖磷酸途径。L-谷氨酸和L-谷氨酰胺共同参与3条代谢通路,其中L-谷氨酸、谷胱甘肽和γ-谷氨酰半胱氨酸参与的代谢通路重要性最大。
Abstract:Ultra-high performance liquid chromatography tandem mass spectrometry (UHPLC-MS/MS) was used for non-targeted metabolomics analysis on ribeye stored at 4 ℃.The results indicated that a total of 914 metabolites were identified from the ribeye,mainly including lipids and lipid molecules,organic acids and derivatives,and organic heterocyclic compounds.Through the random forest method,40 potential differential metabolites were screened out.Cluster analysis and pathway enrichment analysis revealed that these differential metabolites primarily participated in nitrogen metabolism,glutathione metabolism,glyceric acid and dicarboxylic acid metabolism,arginine biosynthesis,pyrimidine metabolism,and pentose phosphate pathway.Lglutamic acid and L-glutamine were jointly involved in three different metabolic pathways,among which the metabolic pathways involving L-glutamic acid,glutathione,and γ-glutamylcysteine were of the greatest importance.
