中文摘要: 为建立黑参浸膏质量控制与评价的有效方法,该文采用高效液相色谱-串联质谱(high performance liquid chromatography-tandem mass spectrometry,HPLC-MS/MS)技术对不同原料的黑参浸膏进行深入分析。黑参浸膏样品经70%甲醇溶液提取,采用Thermo Scientific-C18 (100 mm×2.1 mm,2.6 μm)色谱柱梯度洗脱,电喷雾离子源多反应监测,外标法对21种人参皂苷(如人参皂苷Rg1、Re、Rf、Rb1、Rc等)进行定量分析,并构建HPLC-MS/MS指纹图谱用于鉴别与分析。研究表明,21种人参皂苷在0.2~10.0 μg/mL质量浓度范围内线性关系良好(相关系数R2均大于0.998 6);该方法的重复性、稳定性、精密度和回收率均良好,符合定量分析要求。21种人参皂苷相对标准偏差(relative standard deviation,RSD)为3.1%~7.8%,回收率为88.8%~99.8%。所测样品黑参浸膏中,一批黑参浸膏检出西洋参特征成分拟人参皂苷F11和人参皂苷Rf,其他黑参浸膏均检出人参皂苷Rf。

Abstract:To establish an effective method for quality control and evaluation of black ginseng extract, this study utilized high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) to conduct an in-depth analysis of black ginseng extracts from different raw materials. The black ginseng extract samples were extracted with a 70% methanol solution. Separation was achieved using a Thermo Scientific-C18 column(100 mm×2.1 mm, 2.6 μm) with gradient elution. Quantitative analysis of 21 ginsenosides (ginsenosides Rg1, Re, Rf, Rb1, Rc, etc.) was performed using an electrospray ionization source in multiple reaction monitoring mode with an external standard method. An HPLC-MS/MS fingerprint was also constructed for identification and analysis. The research showed that the 21 ginsenosides exhibited good linear relationships within the concentration range of 0.2-10.0 μg/mL, with all correlation coefficients R2 greater than 0.998 6. The method demonstrated satisfactory repeatability, stability, precision, and recovery rate, meeting the requirements for quantitative analysis. The relative standard deviations (RSD) for the 21 ginsenosides ranged from 3.1% to 7.8%, and the recovery rate was between 88.8% and 99.8%. Among the tested black ginseng extract samples, one batch was found to contain pseudoginsenoside F11 and ginsenoside Rf, which were characteristic components of American ginseng, while ginsenoside Rf was detected in all other black ginseng extracts.
